Abstract

The present invention provides a method of sequencing all or part of a target nucleic acid molecule by determining the sequence of a portion of said nucleic acid molecule in addition to information regarding the position of said portion and in particular provides a new method of sequencing involving the magnification of one or more bases of said bases to aid identification.

         26 . A molecule for magnifying a signal associated with at least one base in a target nucleic acid molecule, comprising:
 an adapter for hybridizing to at least one base in a single strand of a target nucleic acid molecule, wherein the adapter includes at least one complementary base for the at least one base;   a magnifying tag joined to the adapter, wherein the magnifying tag comprises a tag adapted to produce a magnified signal indicative of a characteristic of the at least one complementary base, wherein the characteristic is at least one of a size and a spatial configuration.   
     
     
         27 . The molecule according to  claim 26 , wherein the adapter includes a cleavage site. 
     
     
         28 . The molecule according to  claim 26 , further comprising a link portion adapted to couple to an adjacent adapter hybridized to the single strand. 
     
     
         29 . A kit for magnifying a signal associated with at least one base in a target nucleic acid molecule:
 at least four molecules for magnifying signals associated with respective bases in a target nucleic acid molecule, wherein each of the at least four molecules comprises:   an adapter for hybridizing to a respective base in a single strand of the target nucleic acid molecule, wherein the adapter includes a complementary base for the respective base; and   a magnifying tag joined to the adapter, wherein the magnifying tag comprises a tag adapted to produce a magnified signal indicative of a characteristic of the complementary base, wherein the characteristic is at least one of a size and a spatial configuration,   an enzyme adapted to join adjacent ones of the at least four molecules to form a complimentary strand to the single strand and to join the complimentary strand to the single strand to form a double-strand; and   a nanopore adapted to receive the complimentary strand when separated from the double strand and adapted to generate the magnified signal based on the tag.
