Abstract
The present invention discloses humanised anti-IL-18 antibodies, methods of manufacture, and methods of treatment with said antibodies. Further disclosed are screening methods using for example surface plasmon resonance to identify antibodies with therapeutic potential.

1 . A humanized anti-interleukin-18 antibody comprising
a heavy chain comprising an amino acid sequence selected from the group consisting of: SEQ ID NO: 11, SEQ ID NO: 19, and SEQ ID NO: 23; and a light chain comprising an amino acid sequence selected from the group consisting of: SEQ ID NO: 15 and SEQ ID NO: 31.
2 . A humanized anti-interleukin-18 antibody comprising a heavy chain comprising an amino acid sequence selected from the group consisting of: SEQ ID NO: 11, SEQ ID NO: 19, and SEQ ID NO: 23.
3 . A humanized anti-interleukin-18 antibody comprising a light chain comprising an amino acid sequence selected from the group consisting of: SEQ ID NO: 15 and SEQ ID NO: 31.
4 . The humanized anti-interleukin-18 antibody of claim 1 , wherein said antibody comprises a heavy chain comprising the amino acid sequence shown in SEQ ID NO: 11 and a light chain comprising the amino acid sequence shown in SEQ ID NO: 15.
5 . A method of treating a disease or disorder responsive to neutralization of interleukin-18 in a human in need thereof, said method comprising administering to the human a therapeutically effective amount of an antibody according to claim 1 .

The method of claim 5 , wherein said antibody comprises a heavy chain comprising an amino acid sequence selected from the group consisting of: SEQ ID NO: 9, SEQ ID NO: 17, and SEQ ID NO: 21; and a light chain comprising an amino acid sequence selected from the group consisting of: SEQ ID NO: 13 and SEQ ID NO: 29.

The method of claim 5 , wherein said antibody comprises a heavy chain comprising the amino acid sequence shown in SEQ ID NO: 11 and a light chain comprising the amino acid sequence shown in SEQ ID NO: 15, or a heavy chain comprising the amino acid sequence shown in SEQ ID NO: 11 and a light chain comprising the amino acid sequence shown in SEQ ID NO: 31.

The method of claim 7 , wherein said antibody comprises a heavy chain comprising the amino acid sequence shown in SEQ ID NO: 9 and a light chain comprising the amino acid sequence shown in SEQ ID NO: 13, or a heavy chain comprising the amino acid sequence shown in SEQ ID NO: 9 and a light chain comprising the amino acid sequence shown in SEQ ID NO: 29.
9 . The method of claim 5 , wherein said antibody comprises a heavy chain comprising the amino acid sequence shown in SEQ ID NO: 19 and a light chain comprising the amino acid sequence shown in SEQ ID NO: 15, or a heavy chain comprising the amino acid sequence shown in SEQ ID NO: 19 and a light chain comprising the amino acid sequence shown in SEQ ID NO: 31.

The method of claim 9 , wherein said antibody comprises a heavy chain comprising the amino acid sequence shown in SEQ ID NO: 17 and a light chain comprising the amino acid sequence shown in SEQ ID NO: 13, or a heavy chain comprising the amino acid sequence shown in SEQ ID NO: 17 and a light chain comprising the amino acid sequence shown in SEQ ID NO: 29.

The method of claim 5 , wherein said antibody comprises a heavy chain comprising the amino acid sequence shown in SEQ ID NO: 23 and a light chain comprising the amino acid sequence shown in SEQ ID NO: 15, or a heavy chain comprising the amino acid sequence shown in SEQ ID NO: 23 and a light chain comprising the amino acid sequence shown in SEQ ID NO: 31.

The method of claim 11 , wherein said antibody comprises a heavy chain comprising the amino acid sequence shown in SEQ ID NO: 21 and a light chain comprising the amino acid sequence shown in SEQ ID NO: 13, or a heavy chain comprising the amino acid sequence shown in SEQ ID NO: 21 and a light chain comprising the amino acid sequence shown in SEQ ID NO: 29.
13 . The method of claim 5 , wherein said antibody comprises a heavy chain comprising the amino acid sequence shown in SEQ ID NO: 11 and a light chain comprising the amino acid sequence shown in SEQ ID NO: 15.
14 . The method of claim 13 , wherein said antibody comprises a heavy chain comprising the amino acid sequence shown in SEQ ID NO: 9 and a light chain comprising the amino acid sequence shown in SEQ ID NO: 13.
15 . A method of treating a disease or disorder responsive to neutralization of interleukin-18 in a human in need thereof, said method comprising administering to the human a therapeutically effective amount of an antibody according to claim 2 .
16 . The method of claim 15 , wherein said antibody comprises a heavy chain selected from the group consisting of: SEQ ID NO: 9, SEQ ID NO: 17, and SEQ ID NO: 21.
17 . A method of treating a disease or disorder responsive to neutralization of interleukin-18 in a human in need thereof, said method comprising administering to the human a therapeutically effective amount of an antibody according to claim 3 .
18 . The method of claim 17 , wherein said antibody comprises a light chain selected from the group consisting of: SEQ ID NO: 13 and SEQ ID NO: 29.
19 . A method for the production of the antibody according to claim 1 , said method comprising culturing a host cell transformed or transfected with a vector comprising a polynucleotide encoding said antibody under conditions permissive for the expression of said antibody.
20 . The method of claim 19 , wherein said antibody comprises a heavy chain comprising the amino acid sequence shown in SEQ ID NO: 11 and a light chain comprising the amino acid sequence shown in SEQ ID NO: 15.Cited by (0)
