Abstract

The present invention discloses humanised anti-IL-18 antibodies, methods of manufacture, and methods of treatment with said antibodies. Further disclosed are screening methods using for example surface plasmon resonance to identify antibodies with therapeutic potential.

What is claimed is: 
     
         1 . A humanized anti-interleukin-18 antibody comprising
 a heavy chain comprising an amino acid sequence selected from the group consisting of: SEQ ID NO: 11, SEQ ID NO: 19, and SEQ ID NO: 23; and   a light chain comprising an amino acid sequence selected from the group consisting of: SEQ ID NO: 15 and SEQ ID NO: 31.   
     
     
         2 . A humanized anti-interleukin-18 antibody comprising a heavy chain comprising an amino acid sequence selected from the group consisting of: SEQ ID NO: 11, SEQ ID NO: 19, and SEQ ID NO: 23. 
     
     
         3 . A humanized anti-interleukin-18 antibody comprising a light chain comprising an amino acid sequence selected from the group consisting of: SEQ ID NO: 15 and SEQ ID NO: 31. 
     
     
         4 . The humanized anti-interleukin-18 antibody of  claim 1 , wherein said antibody comprises a heavy chain comprising the amino acid sequence shown in SEQ ID NO: 11 and a light chain comprising the amino acid sequence shown in SEQ ID NO: 15. 
     
     
         5 . A method of treating a disease or disorder responsive to neutralization of interleukin-18 in a human in need thereof, said method comprising administering to the human a therapeutically effective amount of an antibody according to  claim 1 . 
     
     
         6 . The method of  claim 5 , wherein said antibody comprises a heavy chain comprising an amino acid sequence selected from the group consisting of: SEQ ID NO: 9, SEQ ID NO: 17, and SEQ ID NO: 21; and a light chain comprising an amino acid sequence selected from the group consisting of: SEQ ID NO: 13 and SEQ ID NO: 29. 
     
     
         7 . The method of  claim 5 , wherein said antibody comprises a heavy chain comprising the amino acid sequence shown in SEQ ID NO: 11 and a light chain comprising the amino acid sequence shown in SEQ ID NO: 15, or a heavy chain comprising the amino acid sequence shown in SEQ ID NO: 11 and a light chain comprising the amino acid sequence shown in SEQ ID NO: 31. 
     
     
         8 . The method of  claim 7 , wherein said antibody comprises a heavy chain comprising the amino acid sequence shown in SEQ ID NO: 9 and a light chain comprising the amino acid sequence shown in SEQ ID NO: 13, or a heavy chain comprising the amino acid sequence shown in SEQ ID NO: 9 and a light chain comprising the amino acid sequence shown in SEQ ID NO: 29. 
     
     
         9 . The method of  claim 5 , wherein said antibody comprises a heavy chain comprising the amino acid sequence shown in SEQ ID NO: 19 and a light chain comprising the amino acid sequence shown in SEQ ID NO: 15, or a heavy chain comprising the amino acid sequence shown in SEQ ID NO: 19 and a light chain comprising the amino acid sequence shown in SEQ ID NO: 31. 
     
     
         10 . The method of  claim 9 , wherein said antibody comprises a heavy chain comprising the amino acid sequence shown in SEQ ID NO: 17 and a light chain comprising the amino acid sequence shown in SEQ ID NO: 13, or a heavy chain comprising the amino acid sequence shown in SEQ ID NO: 17 and a light chain comprising the amino acid sequence shown in SEQ ID NO: 29. 
     
     
         11 . The method of  claim 5 , wherein said antibody comprises a heavy chain comprising the amino acid sequence shown in SEQ ID NO: 23 and a light chain comprising the amino acid sequence shown in SEQ ID NO: 15, or a heavy chain comprising the amino acid sequence shown in SEQ ID NO: 23 and a light chain comprising the amino acid sequence shown in SEQ ID NO: 31. 
     
     
         12 . The method of  claim 11 , wherein said antibody comprises a heavy chain comprising the amino acid sequence shown in SEQ ID NO: 21 and a light chain comprising the amino acid sequence shown in SEQ ID NO: 13, or a heavy chain comprising the amino acid sequence shown in SEQ ID NO: 21 and a light chain comprising the amino acid sequence shown in SEQ ID NO: 29. 
     
     
         13 . The method of  claim 5 , wherein said antibody comprises a heavy chain comprising the amino acid sequence shown in SEQ ID NO: 11 and a light chain comprising the amino acid sequence shown in SEQ ID NO: 15. 
     
     
         14 . The method of  claim 13 , wherein said antibody comprises a heavy chain comprising the amino acid sequence shown in SEQ ID NO: 9 and a light chain comprising the amino acid sequence shown in SEQ ID NO: 13. 
     
     
         15 . A method of treating a disease or disorder responsive to neutralization of interleukin-18 in a human in need thereof, said method comprising administering to the human a therapeutically effective amount of an antibody according to  claim 2 . 
     
     
         16 . The method of  claim 15 , wherein said antibody comprises a heavy chain selected from the group consisting of: SEQ ID NO: 9, SEQ ID NO: 17, and SEQ ID NO: 21. 
     
     
         17 . A method of treating a disease or disorder responsive to neutralization of interleukin-18 in a human in need thereof, said method comprising administering to the human a therapeutically effective amount of an antibody according to  claim 3 . 
     
     
         18 . The method of  claim 17 , wherein said antibody comprises a light chain selected from the group consisting of: SEQ ID NO: 13 and SEQ ID NO: 29. 
     
     
         19 . A method for the production of the antibody according to  claim 1 , said method comprising culturing a host cell transformed or transfected with a vector comprising a polynucleotide encoding said antibody under conditions permissive for the expression of said antibody. 
     
     
         20 . The method of  claim 19 , wherein said antibody comprises a heavy chain comprising the amino acid sequence shown in SEQ ID NO: 11 and a light chain comprising the amino acid sequence shown in SEQ ID NO: 15.
