Abstract
The present invention relates to the field of biotechnology, in particular to application of a GPR45 gene. The present invention discloses, for the first time, a correlation between GPR45 and obesity and also discloses that obesity may be caused if the GPR45 gene is knocked out or the expression of the GPR45 gene is reduced. Moreover, an obese mouse model is established by adopting a method of blocking the expression of the GPR45 gene for the first time, which is more similar to the mechanism underlying the obesity of human, is thus an ideal model for obesity basis and clinical application researches and can be well applied in screening of drugs for treating obesity.

2 . The application according to claim 1 , characterized in that the animals are mammals.
3 . The application according to claim 1 , characterized in that the application specifically refers to establishment of the obesity animal model by knocking out GPR45 genes in bodies of the animals or suppressing expression of the GPR45 genes in the bodies of the animals.
4 . A method for establishing an obesity animal model, comprising the following step: knocking out GPR45 genes in bodies of animals or suppressing expression of the GPR45 genes in the bodies of the animals to obtain the obesity animal model.
5 . The method according to claim 4 , characterized in that the animals are mammals.
6 . The method according to claim 4 , characterized in that the mammals are mice or rats.
7 . An obesity animal model, established by adopting the method according to claim 4 .
8 . The obesity animal model according to claim 7 , characterized in that the obesity animal model can be further developed to form a hepatic steatosis animal model or a diabetes animal model.
9 . Application of the animal model according to claim 8 in screening of weight-reducing drugs.
10 . A method for screening weight-reducing drug candidates, comprising the following steps: administrating test drug candidates to an obesity animal model established by the method of claim 4 , and comparing with an obesity animal model to which the test drug candidates are not administrated, wherein test compounds which improve or cure obesity after being administrated are drug candidates of weight-reducing drugs.

12 . A method for screening drug candidates or preparations for treating obesity, comprising the following steps:
(1) adding a to-be-tested substance into a system for expressing a GPR45 gene; and (2) detecting a transcription or expression level or the activity of the expression product or the cell activity of the GPR45 gene or GPR45 signal downstream gene, in the system, wherein, compared with an experiment in which the to-be-tested substance is not added, if the to-be-tested substance can promote or improve transcription or expression of the GPR45 gene, or change the transcription or the expression or the expression product activity or the cell activity of GPR45 signal downstream gene accordingly, that the candidate drug can be used as a candidate drug or preparation for treating obesity is indicated.Join the waitlist — get patent alerts
