Abstract

Methods for suppressing IgE-mediated anaphylaxis are provided herein, which include administering to a person in need thereof a combination of at least two therapeutic agents selected from the group consisting of an antihistamine, one or more beta-adrenergic agonists, and one or more tyrosine kinase antagonists. Also provided herein are methods of suppressing IgE-mediated anaphylaxis associated with immunotherapeutic desensitization of a subject, and pharmaceutical compositions for suppressing IgE-mediated anaphylaxis.

Claims

exact text as granted — not AI-modified
1 . A method of suppressing IgE-mediated anaphylaxis comprising administering to a subject in need thereof a combination of least two therapeutic agents selected from the group consisting of imatinib, fostamatinib, and triprolidine.

2 . The method of claim 1 , further comprising administering an antigen or an antibody selected from the group consisting of an anti-IgE monoclonal antibody, an anti-Fc epsilon receptor I alpha (FcεRIα) monoclonal antibody, and combinations thereof.

         3 - 5 . (canceled)

6 . The method of claim 2 , further comprising administering a corticosteroid.

7 . The method of claim 6 , wherein the corticosteroid comprises dexamethasone.

8 . The method of claim 2 , wherein the combination of at least two therapeutic agents is administered prior to administering the antigen or antibody.

9 . The method of claim 1 , wherein the at least two therapeutic agents are administered simultaneously or consecutively.

10 . A method of suppressing IgE-mediated anaphylaxis associated with immunotherapeutic desensitization of a subject in need thereof, the method comprising:
 a) treating the subject with a combination of at least two therapeutic agents selected from the group consisting of imatinib, fostamatinib, and triprolidine; and b) administering to the subject an antigen or an antibody selected from the group consisting of an anti-IgE monoclonal antibody, an anti-Fc epsilon receptor I alpha (FcεRIα) monoclonal antibody, and combinations thereof, wherein the treating step (a) suppresses IgE-mediated anaphylaxis in the subject.

11 . The method of claim 10 , further comprising administering a corticosteroid.

12 . The method of claim 11 , wherein the corticosteroid comprises dexamethasone.

13 . The method of claim 11 , wherein the corticosteroid is administered prior to administering the antigen or antibody.

         14 - 16 . (canceled)

17 . A pharmaceutical composition for suppressing IgE-mediated anaphylaxis comprising:
 (a) a safe and effective amount of at least two therapeutic agents selected from the group consisting of triprolidine, imatinib, and fostamatinib; and (b) one or more pharmaceutically-acceptable excipients.

18 . The pharmaceutical composition of claim 17 , further comprising a corticosteroid.

19 . The pharmaceutical composition of claim 18 , wherein the corticosteroid comprises dexamethasone.

20 . The pharmaceutical composition of claim 17 , further comprising an antigen or an antibody selected from the group consisting of an anti-IgE monoclonal antibody, an anti-Fc epsilon receptor I alpha (FcεRIα) monoclonal antibody, and combinations thereof.

         21 - 22 . (canceled)

23 . The pharmaceutical composition of claim 17 , wherein the at least two therapeutic agents comprise triprolidine, fostamatinib, and imatinib.

         24 - 26 . (canceled)

27 . The method of claim 1 , wherein the combination of at least two therapeutic agents consists of imatinib and fostamatinib.

28 . The method of claim 1 , wherein the combination of at least two therapeutic agents consists of imatinib and triprolidine.

29 . The method of claim 1 , wherein the combination of at least two therapeutic agents consists of fostamatinib and triprolidine.

30 . The method of claim 1 , wherein the combination of at least two therapeutic agents consists of imatinib, fostamatinib, and triprolidine.
