Abstract

Methods are described for phototransferring a compound from a first surface to a second surface. Compounds are described with photocleavable linkers. Compounds attached to a first surface through a photocleavable linker are put in proximity (or contact) with a second surface, and then phototransferred to the second surface upon exposure to electromagnetic radiation. Illuminating the compound with radiation photocleaves the compound from the first surface and transfers the compound to the second surface.

         6 . A method for concentrating a target analyte from a sample using photocleavable binding agents, comprising:
 a) providing:
 i. a sample comprising a target analyte, said sample being a heterogeneous mixture; 
 ii. a plurality of binding agents attached to a surface through a photocleavable linker; and 
 iii. a source of electromagnetic radiation; 
   b) contacting said binding agents with said sample under conditions such that at least a portion of said binding agents bind said target analyte so as to make a complex; and   c) illuminating said complex with radiation from said radiation source under conditions such that said complex, comprising a target analyte bound to said binding agent, is photocleaved from said surface and released into a solution under conditions such that the concentration of said target analyte in said solution is increased relative to said sample.   
     
     
         7 . The method of  claim 6 , wherein said photocleavable linker is a photocleavable biotin. 
     
     
         8 . The method of  claim 6 , wherein said sample is a biological fluid. 
     
     
         9 . The method of  claim 6 , wherein said sample is a body fluid from a mammal. 
     
     
         10 . The method of  claim 9 , wherein said body fluid is selected from the group consisting of blood, serum and plasma. 
     
     
         11 . The method of  claim 6 , wherein said target analyte is a peptide or protein. 
     
     
         12 . The method of  claim 6 , wherein said binding agent is an antibody. 
     
     
         13 . The method of  claim 6 , wherein said binding agent is an aptamer or a modified aptamer. 
     
     
         14 . The method of  claim 6 , wherein said surface is the surface of a bead or microsphere. 
     
     
         15 . The method of  claim 6 , wherein said surface is the surface of a microtiter plate. 
     
     
         16 . The method of  claim 6 , wherein said surface is the surface of a membrane. 
     
     
         17 . The method of  claim 16 , wherein said membrane is comprised of nitrocellulose, cellulose nitrate, cellulose acetate and/or mixed cellulose esters. 
     
     
         18 . A method for purifying a target analyte from a sample using photocleavable binding agents, comprising:
 a) providing:
 i. a sample comprising a target analyte, said sample being a heterogeneous mixture; 
 ii. a plurality of binding agents attached to a surface through a photocleavable linker; and 
 iii. a source of electromagnetic radiation; 
   b) contacting said binding agents with said sample under conditions such that at least a portion of said binding agents bind said target analyte so as to make a complex; and   c) illuminating said complex with radiation from said radiation source under conditions such that said complex, comprising a target analyte bound to said binding agent, is photocleaved from said surface and released into a solution under conditions such that the purity of said target analyte in said solution is increased relative to said sample.   
     
     
         19 . The method of  claim 18 , wherein said photocleavable linker is a photocleavable biotin. 
     
     
         20 . The method of  claim 18 , wherein said sample is a biological fluid. 
     
     
         21 . The method of  claim 18 , wherein said sample is a body fluid from a mammal. 
     
     
         22 . The method of  claim 21 , wherein said body fluid is selected from the group consisting of blood, serum and plasma.

         30 . A method for detecting a target analyte from a sample using photocleavable binding agents, comprising:
 a) providing:
 i. a sample comprising a target analyte, said sample being a heterogeneous mixture; 
 ii. a plurality of binding agents attached to a surface through a photocleavable linker; and 
 iii. a source of electromagnetic radiation; 
   b) contacting said binding agents with said sample under conditions such that at least a portion of said binding agents bind said target analyte so as to make a complex;   c) illuminating said complex with radiation from said radiation source under conditions such that said complex, comprising a target analyte bound to said binding agent, is photocleaved from said surface and released; and   d) detecting said target analyte.   
     
     
         31 . The method of  claim 30 , wherein said photocleavable linker is a photocleavable biotin. 
     
     
         32 . The method of  claim 30 , wherein said sample is a biological fluid. 
     
     
         33 . The method of  claim 30 , wherein said sample is a body fluid from a mammal. 
     
     
         34 . The method of  claim 33 , wherein said body fluid is selected from the group consisting of blood, serum and plasma.
