Abstract

The present invention relates to isolated polypeptides having cellulolytic enhancing activity and isolated polynucleotides encoding the polypeptides. The invention also relates to nucleic acid constructs, vectors, and host cells comprising the polynucleotides as well as methods of producing and using the polypeptides.

A nucleic acid construct or recombinant expression vector comprising an isolated polynucleotide comprising a nucleotide sequence that encodes a GH61 polypeptide having cellulolytic enhancing activity comprising an amino acid sequence having at least 96% sequence identity to amino acids 19 to 349 of SEQ ID NO: 4, wherein the isolated polynucleotide is operably linked to one or more heterologous control sequences that direct the production of the polypeptide in an expression host. 
     
     
         23 . The nucleic acid construct or recombinant expression vector of  claim 22 , wherein the GH61 polypeptide has at least 97% sequence identity to amino acids 19 to 349 of SEQ ID NO: 4. 
     
     
         24 . The nucleic acid construct or recombinant expression vector of  claim 22 , wherein the GH61 polypeptide has at least 98% sequence identity to amino acids 19 to 349 of SEQ ID NO: 4. 
     
     
         25 . The nucleic acid construct or recombinant expression vector of  claim 22 , wherein the GH61 polypeptide has at least 99% sequence identity to amino acids 19 to 349 of SEQ ID NO: 4. 
     
     
         26 . The nucleic acid construct or recombinant expression vector of  claim 22 , wherein the GH61 polypeptide comprises amino acids 19 to 349 of SEQ ID NO: 4.

         30 . A method of producing a GH61 polypeptide having cellulolytic enhancing activity, said method comprising:
 (a) cultivating the recombinant host cell of claim  8  under conditions conducive for production of the polypeptide; and   (b) recovering the polypeptide.   
     
     
         31 . A transgenic plant, plant part or plant cell transformed with the nucleic acid construct or recombinant expression vector of  claim 22 . 
     
     
         32 . A method of producing a GH61 polypeptide having cellulolytic enhancing activity, said method comprising:
 (a) cultivating the transgenic plant or plant cell of  claim 31  under conditions conducive for production of the polypeptide; and   (b) recovering the polypeptide.   
     
     
         33 . A method for degrading a cellulosic material comprising:
 (a) treating the cellulosic material with an enzyme composition comprising a GH61 polypeptide having cellulolytic enhancing activity, wherein the GH61 polypeptide comprises an amino acid sequence having at least 96% sequence identity to amino acids 19 to 349 of SEQ ID NO: 4, and   (b) recovering the degraded cellulosic material.   
     
     
         34 . The method of  claim 33 , wherein the cellulosic material is pretreated. 
     
     
         35 . The method of  claim 33 , wherein the GH61 polypeptide has at least 97% sequence identity to amino acids 19 to 349 of SEQ ID NO: 4. 
     
     
         36 . The method of  claim 33 , wherein the GH61 polypeptide has at least 98% sequence identity to amino acids 19 to 349 of SEQ ID NO: 4. 
     
     
         37 . The method of  claim 33 , wherein the GH61 polypeptide has at least 99% sequence identity to amino acids 19 to 349 of SEQ ID NO: 4. 
     
     
         38 . The method of  claim 33 , wherein the GH61 polypeptide comprises amino acids 19 to 349 of SEQ ID NO: 4. 
     
     
         39 . The method of  claim 33 , wherein the GH61 polypeptide consists of amino acids 19 to 349 of SEQ ID NO: 4, or a fragment thereof having cellulolytic enhancing activity.

         41 . A method for producing a fermentation product, said method comprising:
 (a) saccharifying a cellulosic material with an enzyme composition comprising a GH61 polypeptide having cellulolytic enhancing activity, wherein the GH61 polypeptide comprises an amino acid sequence having at least 96% sequence identity to amino acids 19 to 349 of SEQ ID NO: 4;   (b) fermenting the saccharified cellulosic material with one or more fermenting microorganisms to produce the fermentation product; and   (c) recovering the fermentation product from the fermentation.   
     
     
         42 . The method of  claim 41 , wherein the cellulosic material is pretreated. 
     
     
         43 . The method of  claim 41 , wherein steps (a) and (b) are performed simultaneously. 
     
     
         44 . The method of  claim 41 , wherein the GH61 polypeptide has at least 97% sequence identity to amino acids 19 to 349 of SEQ ID NO: 4. 
     
     
         45 . The method of  claim 41 , wherein the GH61 polypeptide has at least 98% sequence identity to amino acids 19 to 349 of SEQ ID NO: 4. 
     
     
         46 . The method of  claim 41 , wherein the GH61 polypeptide has at least 99% sequence identity to amino acids 19 to 349 of SEQ ID NO: 4. 
     
     
         47 . The method of  claim 41 , wherein the GH61 polypeptide comprises amino acids 19 to 349 of SEQ ID NO: 4.

         50 . A method of fermenting a cellulosic material comprising:
 (a) fermenting the cellulosic material with one or more fermenting microorganisms, wherein the cellulosic material is saccharified with an enzyme composition comprising a GH61 polypeptide having cellulolytic enhancing activity, wherein the GH61 polypeptide comprises an amino acid sequence having at least 96% sequence identity to amino acids 19 to 349 of SEQ ID NO: 4, and wherein the fermenting of the cellulosic material produces a fermentation product, and   (b) recovering the fermentation product from the fermentation.   
     
     
         51 . The method of  claim 29 , wherein the cellulosic material is pretreated. 
     
     
         52 . The method of  claim 29 , further comprising covering the fermentation product. 
     
     
         53 . The method of  claim 29 , wherein the GH61 polypeptide has at least 97% sequence identity to amino acids 19 to 349 of SEQ ID NO: 4. 
     
     
         54 . The method of  claim 29 , wherein the GH61 polypeptide has at least 98% sequence identity to amino acids 19 to 349 of SEQ ID NO: 4.
