Abstract
This document provides methods and materials involved obtaining induced pluripotent stem (iPS) cells. For example, methods and materials for increasing the efficiency for making iPS cells as well as methods and materials for selecting iPS cells are provided.
Claims
exact text as granted — not AI-modified1 - 6 . (canceled)
7 . A method for identifying induced pluripotent stem cells within a sample comprising a mixture of somatic cells and said induced pluripotent stem cells, wherein said method comprises:
(a) contacting said sample with a marker of glycolysis, wherein said marker labels said induced pluripotent stem cells to a greater extent than said somatic cells, and (b) obtaining said induced pluripotent stem cells from said mixture based at least in part on an increased level of said marker being associated with said induced pluripotent stem cells.
8 . The method of claim 7 , wherein said sample is an in vitro culture sample.
9 . The method of claim 7 , wherein said somatic cells are fibroblasts.
10 . The method of claim 7 , wherein said induced pluripotent stem cells were produced from said somatic cells by the expression of stemness transcription factors.
11 - 12 . (canceled)
13 . The method of claim 7 , wherein said induced pluripotent stem cells were produced in the presence of an effective amount of an agent that promotes or aids glycolysis.
14 . The method of claim 13 , wherein said agent is glucose.
15 . The method of claim 13 , wherein said agent is glucose, and said effective amount is between 5 mM and 25 mM.
16 . The method of claim 7 , wherein said marker is lactate.
