Abstract

The present invention relates to isolated polypeptides having cellulolytic enhancing activity and isolated polynucleotides encoding the polypeptides. The invention also relates to nucleic acid constructs, vectors, and host cells comprising the polynucleotides as well as methods of producing and using the polypeptides.

         21 . A process for degrading a cellulosic material, said process comprising: treating the cellulosic material with an enzyme composition comprising a GH61 polypeptide having cellulolytic enhancing activity, wherein the GH61 polypeptide is selected from the group consisting of:
 (a) a GH61 polypeptide having at least 90% sequence identity to the mature polypeptide of SEQ ID NO: 4, SEQ ID NO: 6, SEQ ID NO: 8, SEQ ID NO: 10, or SEQ ID NO: 14, or at least 95% sequence identity to the mature polypeptide of SEQ ID NO: 12;   (b) a GH61 polypeptide encoded by a polynucleotide having at least 95% sequence identity to the mature polypeptide coding sequence of SEQ ID NO: 3, SEQ ID NO: 5, SEQ ID NO: 7, SEQ ID NO: 9, SEQ ID NO: 11, or SEQ ID NO: 13, or the cDNA sequence of SEQ ID NO: 5, SEQ ID NO: 11, or SEQ ID NO: 13;   (c) a GH61 polypeptide comprising the mature polypeptide of SEQ ID NO: 4, SEQ ID NO: 6, SEQ ID NO: 8, SEQ ID NO: 10, SEQ ID NO: 12, or SEQ ID NO: 14; or a fragment thereof, wherein the fragment has cellulolytic enhancing activity; and   (d) a GH61 polypeptide encoded by a polynucleotide comprising the mature polypeptide coding sequence of SEQ ID NO: 3, SEQ ID NO: 5, SEQ ID NO: 7, SEQ ID NO: 9, SEQ ID NO: 11, or SEQ ID NO: 13.   
     
     
         22 . The process of  claim 21 , wherein the GH61 polypeptide comprises the mature polypeptide of SEQ ID NO: 4, SEQ ID NO: 6, SEQ ID NO: 8, SEQ ID NO: 10, or SEQ ID NO: 14. 
     
     
         23 . The process of  claim 21 , wherein the GH61 polypeptide is encoded by a polynucleotide comprising the mature polypeptide coding sequence of SEQ ID NO: 3, SEQ ID NO: 5, SEQ ID NO: 7, SEQ ID NO: 9, SEQ ID NO: 11, or SEQ ID NO: 13. 
     
     
         24 . The process of  claim 21 , wherein the cellulosic material is pretreated. 
     
     
         25 . The process of  claim 21 , wherein the enzyme composition further comprises one or more enzymes selected from the group consisting of a cellulase, a GH61 polypeptide having cellulolytic enhancing activity, a hemicellulase, an esterase, an expansin, a laccase, a ligninolytic enzyme, a pectinase, a peroxidase, a protease, and a swollenin. 
     
     
         26 . The process of  claim 21 , further comprising recovering the degraded cellulosic material. 
     
     
         27 . The process of  claim 26 , wherein the degraded cellulosic material is a sugar. 
     
     
         28 . The process of  claim 27 , wherein the sugar is selected from the group consisting of glucose, xylose, mannose, galactose, and arabinose. 
     
     
         29 . A process for producing a fermentation product, said process comprising:
 (a) saccharifying a cellulosic material with an enzyme composition comprising a GH61 polypeptide having cellulolytic enhancing activity, wherein the GH61 polypeptide is selected from the group consisting of:   (1) a GH61 polypeptide having at least 90% sequence identity to the mature polypeptide of SEQ ID NO: 4, SEQ ID NO: 6, SEQ ID NO: 8, SEQ ID NO: 10, or SEQ ID NO: 14, or at least 95% sequence identity to the mature polypeptide of SEQ ID NO: 12;   (2) a GH61 polypeptide encoded by a polynucleotide having at least 95% sequence identity to the mature polypeptide coding sequence of SEQ ID NO: 3, SEQ ID NO: 5, SEQ ID NO: 7, SEQ ID NO: 9, SEQ ID NO: 11, or SEQ ID NO: 13, or the cDNA sequence of SEQ ID NO: 5, SEQ ID NO: 11, or SEQ ID NO: 13;   (3) a GH61 polypeptide comprising the mature polypeptide of SEQ ID NO: 4, SEQ ID NO: 6, SEQ ID NO: 8, SEQ ID NO: 10, SEQ ID NO: 12, or SEQ ID NO: 14; ora fragment thereof, wherein the fragment has cellulolytic enhancing activity; and   (4) a GH61 polypeptide encoded by a polynucleotide comprising the mature polypeptide coding sequence of SEQ ID NO: 3, SEQ ID NO: 5, SEQ ID NO: 7, SEQ ID NO: 9, SEQ ID NO: 11, or SEQ ID NO: 13;   (b) fermenting the saccharified cellulosic material with one or more fermenting microorganisms to produce the fermentation product; and   (c) recovering the fermentation product from the fermentation.   
     
     
         30 . The process of  claim 29 , wherein the GH61 polypeptide comprises the mature polypeptide of SEQ ID NO: 4, SEQ ID NO: 6, SEQ ID NO: 8, SEQ ID NO: 10, or SEQ ID NO: 14. 
     
     
         31 . The process of  claim 29 , wherein the GH61 polypeptide is encoded by a polynucleotide comprising the mature polypeptide coding sequence of SEQ ID NO: 3, SEQ ID NO: 5, SEQ ID NO: 7, SEQ ID NO: 9, SEQ ID NO: 11, or SEQ ID NO: 13. 
     
     
         32 . The process of  claim 29 , wherein the cellulosic material is pretreated. 
     
     
         33 . The process of  claim 29 , wherein the enzyme composition further comprises one or more enzymes selected from the group consisting of a cellulase, a GH61 polypeptide having cellulolytic enhancing activity, a hemicellulase, an esterase, an expansin, a laccase, a ligninolytic enzyme, a pectinase, a peroxidase, a protease, and a swollenin. 
     
     
         34 . The process of  claim 29 , wherein steps (a) and (b) are performed simultaneously in a simultaneous saccharification and fermentation. 
     
     
         35 . The process of  claim 29 , wherein the fermentation product is an alcohol, an alkane, a cycloalkane, an alkene, an amino acid, a gas, isoprene, a ketone, an organic acid, or polyketide. 
     
     
         36 . A process of fermenting a cellulosic material, said process comprising: fermenting the cellulosic material with one or more fermenting microorganisms, wherein the cellulosic material is saccharified with an enzyme composition comprising a GH61 polypeptide having cellulolytic enhancing activity, and wherein the GH61 polypeptide is selected from the group consisting of:
 (a) a GH61 polypeptide having at least 90% sequence identity to the mature polypeptide of SEQ ID NO: 4, SEQ ID NO: 6, SEQ ID NO: 8, SEQ ID NO: 10, or SEQ ID NO: 14, or at least 95% sequence identity to the mature polypeptide of SEQ ID NO: 12;   (b) a GH61 polypeptide encoded by a polynucleotide having at least 95% sequence identity to the mature polypeptide coding sequence of SEQ ID NO: 3, SEQ ID NO: 5, SEQ ID NO: 7, SEQ ID NO: 9, SEQ ID NO: 11, or SEQ ID NO: 13, or the cDNA sequence of SEQ ID NO: 5, SEQ ID NO: 11, or SEQ ID NO: 13;   (c) a GH61 polypeptide comprising the mature polypeptide of SEQ ID NO: 4, SEQ ID NO: 6, SEQ ID NO: 8, SEQ ID NO: 10, SEQ ID NO: 12, or SEQ ID NO: 14; or a fragment thereof, wherein the fragment has cellulolytic enhancing activity; and   (d) a GH61 polypeptide encoded by a polynucleotide comprising the mature polypeptide coding sequence of SEQ ID NO: 3, SEQ ID NO: 5, SEQ ID NO: 7, SEQ ID NO: 9, SEQ ID NO: 11, or SEQ ID NO: 13.   
     
     
         37 . The process of  claim 36 , wherein the GH61 polypeptide comprises the mature polypeptide of SEQ ID NO: 4, SEQ ID NO: 6, SEQ ID NO: 8, SEQ ID NO: 10, or SEQ ID NO: 14. 
     
     
         38 . The process of  claim 36 , wherein the GH61 polypeptide is encoded by a polynucleotide comprising the mature polypeptide coding sequence of SEQ ID NO: 3, SEQ ID NO: 5, SEQ ID NO: 7, SEQ ID NO: 9, SEQ ID NO: 11, or SEQ ID NO: 13. 
     
     
         39 . The process of  claim 36 , wherein the fermenting of the cellulosic material produces a fermentation product. 
     
     
         40 . The process of  claim 39 , further comprising recovering the fermentation product from the fermentation. 
     
     
         41 . The process of  claim 36 , wherein the cellulosic material is pretreated before saccharification. 
     
     
         42 . The process of  claim 36 , wherein the enzyme composition further comprises one or more enzymes selected from the group consisting of a cellulase, a GH61 polypeptide having cellulolytic enhancing activity, a hemicellulase, an esterase, an expansin, a laccase, a ligninolytic enzyme, a pectinase, a peroxidase, a protease, and a swollenin. 
     
     
         43 . The process of  claim 39 , wherein the fermentation product is an alcohol, an alkane, a cycloalkane, an alkene, an amino acid, a gas, isoprene, a ketone, an organic acid, or polyketide.
