Abstract
Disclosed herein are compounds, pharmaceutical compositions, and methods for the treatment of diseases such as inflammation and/or pain, comprising the administration of a therapeutically effective amount of a non-steroidal, non-opioid compound having GI-/renal-/platelet-sparing activities, and wide safety margins with no appreciable inhibition of either form of cyclooxygenase.

1 . A method of treatment of pain or inflammation in a patient with a history of peptic ulcer disease or at increased risk of gastrointestinal bleeding/ulceration/obstruction and in need of such treatment, comprising administering a therapeutically effective amount of methylglyoxal-bisguanylhydrazone (MGBG), wherein the therapeutically effective amount of MGBG does not inhibit cyclooxygenase and is not an opioid or a steroid, and is gastrointestinal-, platelet-, and renal-sparing.
2 . The method as recited in claim 2 , wherein the compound is formulated for oral delivery.
3 . The method as recited in claim 3 , wherein the compound is formulated for delayed-release oral delivery to the small bowel.
4 . The method as recited in claim 1 , wherein non-steroidal anti-inflammatory drugs (NSAIDs) are not advisable or contraindicated in the patient.
5 . The method as recited in claim 1 , wherein the patient has a bleeding or clotting disorder.
6 . The method as recited in claim 1 , wherein the patient has an increased risk for bleeding.
7 . The method as recited in claim 1 , wherein the patient has a gastrointestinal disorder.
8 . The method as recited in claim 1 , wherein the patient has a renal disorder.
9 . The method as recited in claim 1 , wherein the patient has a disorder in which sodium/fluid retention is undesirable.
10 . The method as recited in claim 1 , wherein the patient is taking an anti-coagulant drug.
11 . The method as recited in claim 1 , wherein the patient has a renal disease, congestive heart failure, or cirrhosis, or is volume-depleted or dehydrated.
12 . The method as recited in claim 1 , wherein the patient has cirrhosis or thrombocytopenia.
13 . The method as recited in claim 1 , wherein the patient is pre-operative for surgery.
14 . The method as recited in claim 1 , wherein the patient has a history of peptic ulcer disease or at increased risk of gastrointestinal bleeding/ulceration/obstruction.Cited by (0)
