Abstract
This invention relates to amphiphile-polymer particles, compositions, methods of making, and methods of use thereof.

1 . A particle comprising:
a water-insoluble polymeric core; and a payload and a first amphiphile within the core.
2 . The particle of claim 1 , further comprising a shell comprising a second amphiphile surrounding the water-insoluble polymeric core.
3 . A particle prepared by a process comprising:
obtaining a first solution comprising a water-insoluble polymer, a payload and a first amphiphile in a water-miscible solvent; mixing the first solution with an aqueous second solution to form an aqueous composition comprising a particle comprising a water-insoluble polymeric core comprising the water-insoluble polymer; the payload; and the first amphiphile.
4 . The particle of claim 3 , wherein the process further comprises mixing the aqueous composition with a solution comprising a second amphiphile to form a shell surrounding the core.
5 . The particle of any one of claims 1 - 4 , wherein the payload and the first amphiphile have complementary electrical charges.
6 . 8 . The particle of claim 7 , wherein the water-insoluble polymeric core comprises {poly[(9,9-di-n-octylfluorenyl-2,7-diyl)-alt-(benzo[2,1,3]thiadiazol-4,8-diyl)]}.
9 . The particle of claim 7 , wherein the water-insoluble polymeric core comprises {poly[2,6-(4,4-bis-(2-ethylhexyl)-4H-cyclopenta [2,1-b;3,4-b′]dithiophene)-alt-4,7(2,1,3-benzothiadiazole)]}.
10 . The particle of any one of claims 1 - 5 , wherein the water-insoluble polymeric core comprises one or more polymers selected from polylactic acids, polyglycolic acids, and copolymers of lactic acid and glycolic acid.
11 . The particle of claim 10 , wherein the water-insoluble polymeric core comprises one or more copolymers of lactic acid and glycolic acid.
12 . The particle of claim 11 , comprising a copolymer of lactic acid and glycolic acid that comprises lactic acid and glycolic acid monomer units in a ratio of about 1:1.
13 . The particle of any one of claims 1 - 12 , wherein the first amphiphile is cationic.
14 . The particle of any one of claims 1 - 12 , wherein the first amphiphile is anionic.
15 . The particle of any one of claims 1 - 12 , wherein the first amphiphile comprises one or more compounds selected from the group consisting of: lecithin, amino dendrimers, PEG-phospholipids, PEG-ceramides, anionic lipids, and cationic lipids.
16 . The particle of claim 15 , wherein the first amphiphile comprises an amino dendrimer.
17 . The particle of claim 15 , wherein the first amphiphile comprises a cationic lipid.

The particle of any one of claims 1 - 13 , wherein the first amphiphile comprises one or more compounds selected from the group consisting of: G0-C14 dendrimer; ethylenediamine branched polyethylenimine; polypropylenimine tetramine dendrimer, generation 1; 3β-[N—(N′,N′-dimethylaminoethane)-carbamoyl]cholesterol; 1,2-dioleoyl-3-trimethylammonium-propane; 1-oleoyl-2-[6-[(7-nitro-2-1,3-benzoxadiazol-4-yl)amino]hexanoyl]-3-trimethylammonium propane; 1,2-dimyristoyl-3-trimethylammonium-propane; 1,2-dipalmitoyl-3-trimethylammonium-propane, 1,2-stearoyl-3-trimethylammonium-propane; 1,2-di-O-octadecenyl-3-trimethylammonium propane.
20 . The particle of any one of claims 1 - 13 , wherein the first amphiphile comprises G0-C14 dendrimer.
21 . The particle of any one of claims 1 - 13 , wherein the first amphiphile comprises polypropylenimine tetramine dendrimer, generation 1.
22 . The particle of any one of claims 1 - 13 , wherein the first amphiphile comprises ethylenediamine branched polyethylenimine.
23 . The particle of any one of claims 1 - 13 , wherein the first amphiphile comprises 1,2-dioleoyl-3-trimethylammonium-propane.
24 . The particle of any one of claims 1 - 13 , wherein the first amphiphile comprises 1,2-di-O-octadecenyl-3-trimethylammonium propane.

67 . A method of preparing a particle according to claim 1 comprising:
obtaining a first solution comprising a water-insoluble polymer, a payload and a first amphiphile in a water-miscible solvent;
mixing the first solution with an aqueous second solution to form an aqueous composition comprising a particle according to claim 1 .
68 . The method of claim 67 , further comprising mixing the aqueous composition with a solution comprising a second amphiphile to form a shell surrounding the core.
69 . The method of claim 67 or 68 , wherein the payload and the first amphiphile have complementary electrical charges.
