Abstract

Provided herein are methods and compositions for cardiac therapy. Such compositions include extracellular-matrix (ECM)-based products that can be used to support tissue repair. The compositions can be used for various purposes. In some cases, they can be introduced into a subject in order to preserve and/or repair damaged heart tissue.

Claims

exact text as granted — not AI-modified
.- 46 . (canceled)

. A composition comprising digested, decellularized extracellular matrix derived from tissue from multiple donors, wherein the extracellular matrix comprises material of molecular weight of less than 300 kDa, wherein the extracellular matrix comprises glycosaminoglycan at a concentration of between about 12.5 μg per mL to about 200 μg per mL of the composition, and wherein the composition is injectable through a catheter having an inner diameter of 25 G or smaller.

. The composition of claim 47 , wherein the composition comprises at least 80% extracellular matrix.

. The composition of claim 48 , wherein the composition comprises at least 95% extracellular matrix.

. The composition of claim 47 , wherein the digested, decellularized extracellular matrix is treated with least one digestive enzyme.

. The composition of claim 50 , wherein the at least one digestive enzyme comprises pepsin.

. The composition of claim 50 , wherein the at least one digestive enzyme comprises a matrix metalloproteinase.

. The composition of claim 50 , wherein the at least one digestive enzyme cleaves the matrix such that the composition gels at greater than 20° C.

. The composition of claim 50 , wherein the at least one digestive enzyme cleaves the matrix such that the composition gels in less than 30 minutes.

. The composition of claim 47 , wherein the digested, decellularized extracellular matrix is lyophilized.

. The composition of claim 55 , wherein the lyophilized, digested, decellularized extracellular matrix is miscible in a liquid.

. The composition of claim 55 , wherein the lyophilized, digested, decellularized extracellular matrix is solubilized with a liquid.

. The composition of claim 55 , wherein the lyophilized, digested, decellularized extracellular matrix is storage stable for at least 6 months at a temperature of less than 25° C.

. The composition of claim 47 , wherein the composition further comprises a synthetic polymer or a naturally derived polymer.

. The composition of claim 47 , where in the tissue is cardiac tissue.

. The composition of claim 47 , wherein the multiple donors comprise pigs.

. The composition of claim 47 , wherein the multiple donors comprise humans.

. A method for repairing cardiac tissue comprising injecting in a subject the composition of claim 47 .

. The method of claim 63 , wherein the composition is injected onto a repair site on the cardiac tissue.

. The method of claim 63 , wherein the composition is injected using a transendocardial catheter.

. The method of claim 63 , wherein the composition is injected using a transcoronary catheter.
