Treatment for dopaminergic disorders

Abstract

The present invention provides systems, compositions and methods for treatment of dopaminergic disorders (e.g., Parkinson's disease) using the combination of a first compound that inhibits a voltage-gated calcium channel of the type Ca v 1.3 (e.g., a dihydropyridine such as isradipine), and a second compound that is monoamine oxidase inhibitor and/or is a nitric oxide synthase inhibitor (e.g., rasagiline or derivative thereof).

         21 . A method of treatment for Parkinson's disease comprising: administering to a human subject having Parkinson's disease:
 i) a first compound that comprises isradipine, wherein said first compound is in a controlled release formulation, and   ii) a second compound that comprises rasagiline, and   wherein said administering provides about 0.5 mg to about 2 mg of said rasagiline per day to said subject, and   wherein said administering is repeated for multiple days.   
     
     
         22 . The method of  claim 21 , wherein said administering provides about 1 mg of said rasagiline per day to said subject. 
     
     
         23 . The method of  claim 21 , wherein said administering provides about 10 mg of said isradipine per day to said subject. 
     
     
         24 . The method of  claim 21 , wherein said administering provides 2-90 mg of said isradipine per day. 
     
     
         25 . The method of  claim 21 , wherein said first and second compounds are part of a single composition. 
     
     
         26 . The method of  claim 25 , wherein said single composition is in pill or capsule form. 
     
     
         27 . A composition comprising:
 i) a first compound that comprises isradipine, wherein said first compound is in a controlled release formulation, and   ii) a second compound that comprises rasagiline present in said composition between about 0.5 mg and 2 mg.   
     
     
         28 . The composition of  claim 27 , wherein about 1 mg of said rasagiline is present in said composition. 
     
     
         29 . The composition of  claim 27 , wherein about 10 mg of said isradipine is present in said composition. 
     
     
         30 . The composition of  claim 27 , wherein about 2-90 mg of said isradipine is present in said composition. 
     
     
         31 . The composition of  claim 27 , wherein said composition is in pill or capsule form. 
     
     
         32 . A kit comprising:
 i) a first compound that comprises isradipine, wherein said first compound is in a controlled release formulation, and   ii) a second compound that comprises rasagiline present in said kit between about 0.5 mg and 2 mg.   
     
     
         33 . The kit of  claim 32 , wherein about 1 mg of said rasagiline is present in said kit. 
     
     
         34 . The kit of  claim 32 , wherein about 10 mg of said isradipine is present in said kit. 
     
     
         35 . The kit of  claim 32 , wherein about 2-90 mg of said isradipine is present in said kit. 
     
     
         36 . The kit of  claim 32 , further comprising: iii) a container, wherein said first and second compounds are inside said container. 
     
     
         37 . The kit of  claim 32 , wherein said first and second compounds are in pill or capsule form.
