Abstract

Disclosed are methods and products for improving the quality of sperm, methods for treating subjects to improve sperm quality or improve fertility, and use of sperm with improved quality in assisted reproductive technologies, including exposing the sperm to BGP-15 and/or a derivative thereof, such as BGP-15, propanolol, bimoclomol, arimoclomal, NG94, iroxanadine, and/or a pharmaceutically acceptable derivative, prodrug, solvate, salt, tautomer, stereoisomer, and/or racemate thereof.

1 . A method of improving quality of sperm, the method comprising exposing the sperm to BGP-15 and/or a derivative thereof and thereby improving the quality of the sperm. 
     
     
         2 . The method according to  claim 1 , wherein the BGP-15 and/or a derivative thereof comprises one or more of BGP-15, propanolol, bimoclomol, arimoclomal, NG-94, iroxanadine, and/or a pharmaceutically acceptable derivative, prodrug, solvate, salt, tautomer, stereoisomer, and/or racemate of any of the aforementioned. 
     
     
         3 . The method according to  claim 1 , wherein the BGP-15 and/or a derivative thereof comprises BGP-15. 
     
     
         4 . The method according to  claim 1 , wherein the quality of the sperm comprises fertilization capacity. 
     
     
         5 . The method according to  claim 4 , wherein the fertilization capacity comprises ability to fertilize an oocyte and/or developmental competence of an embryo produced from the sperm. 
     
     
         6 . The method according to  claim 1 , wherein the sperm is a sperm from an aged donor, and/or an obese or overweight donor 
     
     
         7 . The method according to  claim 1 , wherein the sperm is in vitro. 
     
     
         8 . The method according to  claim 7 , wherein the sperm is exposed to the BGP-15 and/or a derivative thereof for a time of one hour or less. 
     
     
         9 . The method according to  claim 1 , wherein the sperm is in vivo in a subject. 
     
     
         10 . The method according to  claim 9 , wherein the exposing of the sperm to the BGP-15 and/or a derivative thereof comprises administering the BGP-15 and/or a derivative thereof to a subject. 
     
     
         11 . The method according to  claim 9 , wherein the subject comprises sperm with a reduced fertilization capacity. 
     
     
         12 . The method according to  claim 9 , wherein the subject is an aged subject or an obese or overweight subject. 
     
     
         13 . Isolated sperm produced by exposing the sperm to BGP-15 and/or a derivative thereof according to  claim 1 . 
     
     
         14 . A method of treating a subject to improve sperm quality, the method comprising administering to the subject to an effective amount of BGP-15 and/or a derivative thereof and thereby improving sperm quality in the subject. 
     
     
         15 . A method of treating reduced sperm quality in a subject, the method comprising administering to the subject an effective amount of BGP-15 and/or a derivative thereof and thereby treating the subject. 
     
     
         16 . Isolated sperm obtained from a subject administered with BGP-15 and/or a derivative thereof according to the method of  claim 14 .

         18 . A pharmaceutical composition for improving sperm quality, the composition comprising an effective amount of BGP-15 and/or a derivative thereof. 
     
     
         19 . A method of treating a subject to improve sperm quality, the method comprising administering to the subject a pharmaceutical composition according to  claim 18 . 
     
     
         20 . A method of improving developmental competence of an embryo produced by a sperm fertilising an oocyte, the method comprising exposing the sperm to BGP-15 and/or a derivative thereof and thereby improving the developmental competence of the embryo.

The method according to  claim 20 , wherein the BGP-15 and/or a derivative thereof comprises one or more of BGP-15, propanolol, bimoclomol, arimoclomal, NG-94, iroxanadine, and/or a pharmaceutically acceptable derivative, prodrug, solvate, salt, tautomer, stereoisomer, or racemate of any of the aforementioned. 
     
     
         22 . The method according to  claim 20 , wherein the BGP-15 and/or a derivative thereof comprises BGP-15. 
     
     
         23 . The method according to  claim 20 , wherein the sperm is a sperm from an aged donor and/or an obese or overweight donor. 
     
     
         24 . The method according to  claim 20 , wherein the method comprises exposing the sperm to the BGP-15 and/or a derivative thereof in vitro. 
     
     
         25 . The method according to  claim 20 , wherein the method comprises exposing the sperm to the BGP-15 and/or a derivative thereof in vivo in a subject. 
     
     
         26 . The method according to  claim 25 , wherein the method comprises administering the BGP-15 and/or a derivative thereof to the subject. 
     
     
         27 . A method of assisted reproduction using sperm, the method comprising exposing the sperm to BGP-15 and/or a derivative thereof and using the sperm exposed to the BGP-15 and/or a derivative thereof in the method of assisted reproduction. 
     
     
         28 . The method according to  claim 27 , wherein the sperm is a sperm from an aged donor and/or an obese or overweight donor. 
     
     
         29 . The method according to  claim 27 , wherein the method of assisted reproduction comprises in vitro fertilization. 
     
     
         30 . The method according to  claim 27 , wherein the method of assisted reproduction comprises artificial insemination. 
     
     
         31 . An animal produced using an assisted reproduction method according to  claim 27 . 
     
     
         32 . An in vitro fertilization method, the method comprising:
 exposing sperm in vitro to BGP-15 and/or a derivative thereof; and
 fertilizing an oocyte with the sperm exposed to the BGP-15 and/or a derivative thereof. 
   
     
     
         33 . A kit or product for improving quality of sperm, the kit or product comprising BGP-15 and/or a derivative thereof for exposure to the sperm.

         35 . A kit or product for performing a method according to  claim 1 . 
     
     
         36 . An in vitro fertilization medium for sperm, the medium comprising BGP-15 and/or a derivative thereof.

         38 . A method of freezing sperm, the method comprising exposing the sperm to BGP-15 and/or a derivative thereof at one more of before, during and/or after freezing of the sperm. 
     
     
         39 . A sperm freezing medium, the medium comprising BGP-15 and/or a derivative thereof. 
     
     
         40 . A method of screening for an agent to improve sperm quality, the method comprising determining the ability of a derivative of BGP-15 to improve quality of sperm and identifying the derivative as an agent to improve sperm quality. 
     
     
         41 . The method according to  claim 40 , wherein the method comprises determining the ability of the derivative in vitro to improve the quality of the sperm. 
     
     
         42 . The method according to  claim 40 , wherein the method comprises determining the ability of the derivative in vivo to improve the quality of the sperm. 
     
     
         43 . The method according to  claim 40 , wherein the derivative comprises a derivative of one of propanolol, bimoclomol, arimoclomal, NG-94, and iroxanadine.
