Claims

Abstract

The subject invention relates to isolated proteins, particularly monoclonal antibodies, which bind and neutralize RGM A protein. Specifically, these antibodies have the ability to inhibit the binding of RGM A to its receptor and/or coreceptors. These antibodies or portions thereof of the invention are useful for detecting RGM A and for inhibiting RGM A activity, for example in a human suffering from a disorder including but nor limited to multiple sclerosis, mammalian brain trauma, spinal cord injury, stroke, neurodegenerative diseases, and schizophrenia.

An isolated monoclonal anti-Repulsive Guidance Molecule A (RGMA) antibody comprising a heavy chain variable region comprising a complementarity determining region(CDR)1 with the amino acid sequence set forth in SEQ ID NO:57, a CDR2 with the amino acid sequence set forth in SEQ ID NO:58, a CDR3 with the amino acid sequence set forth in SEQ ID NO:59; and a light chain variable region comprising a CDR1 with the amino acid sequence set forth in SEQ ID NO:60, a CDR2 with the amino acid sequence set forth in SEQ ID NO:61, a CDR3 with the amino acid sequence set forth in SEQ ID NO:62 
     
     
         93 . The isolated monoclonal anti-RGMA antibody of  claim 92 , wherein heavy chain variable region has at least 90% amino acid sequence identity with the sequence of SEQ ID NO.:34. 
     
     
         94 . The isolated monoclonal anti-RGMA antibody of  claim 92 , wherein light chain variable region has at least 90% amino acid sequence identity with the sequence of SEQ ID NO.:10. 
     
     
         95 . The isolated monoclonal anti-RGMA antibody of  claim 92 , wherein heavy chain variable region comprises the amino acid sequence set forth in SEQ ID NO:34; and the light chain variable region comprises the amino acid sequence set forth in SEQ ID NO:10. 
     
     
         96 . The isolated monoclonal anti-RGMA antibody of  claim 92 , wherein heavy chain variable region consists of the amino acid sequence set forth in SEQ ID NO:34; and the light chain variable region consists of the amino acid sequence set forth in SEQ ID NO:10.
