Abstract

The present invention relates to antibodies, or antigen-binding fragments thereof, directed against human leukocyte antigen-G (HLA-G) protein and raised against an immunogenic peptide derived from the α3 domain of HLA-G protein. The invention further relates to the immunogenic peptide, and methods for producing said anti-HLA-G specific antibodies.

         6 . An anti-HLA-G antibody or an antigen-binding fragment thereof, wherein the antibody or antigen-binding fragment comprises:
 (a) a heavy chain variable region (VH), which comprises a heavy chain complementary determining region 1 (HC CDR1) of SEQ ID NO: 23, a heavy chain complementary determining region 2 (HC CDR2) of SEQ ID NO: 25, and a heavy chain complementary determining region 3 (HC CDR3) of SEQ ID NO: 27; and   (b) a light chain variable region (VL), which comprises a light chain complementary determining region 1 (LC CDR1) of SEQ ID NO: 15, a light chain complementary determining region 2 (LC CDR2) of sequence KVS and a light chain complementary determining region 3 (LC CDR3) of SEQ ID NO: 18.   
     
     
         7 . The antibody or antigen-binding fragment of  claim 6 , wherein the VH comprises SEQ ID NO: 67 and the VL comprises SEQ ID NO: 65. 
     
     
         8 . The antibody or antigen-binding fragment of  claim 6 , wherein the antibody or antigen-binding fragment is chimeric or humanized. 
     
     
         9 . A nucleic acid comprising a nucleotide sequence encoding an antibody heavy chain variable region (VH), an antibody light chain variable region (VL) or both, of the antibody or antigen-binding fragment of claim  1 . 
     
     
         10 . A vector, comprising the nucleic acid of  claim 9 . 
     
     
         11 . A host cell comprising the vector of  claim 10 . 
     
     
         12 . A method for producing an anti-HLA-G monoclonal antibody, comprising culturing the host cell of  claim 11  under conditions allowing for expression of the antibody. 
     
     
         13 . A conjugate comprising the antibody or antigen-binding fragment of claim  1 , linked to a cytotoxic agent. 
     
     
         14 . A method for treating a cancer or a viral infection, the method comprising administering to the subject the antibody or antigen-binding fragment of claim  1 .
