Abstract

The present disclosure provides humanized and affinity matured antibodies (mAbs) and fragments thereof that specifically bind to CD79 with high affinity. The anti-CD79 mAbs and fragments thereof can be used to treat antibody-associated conditions, including autoimmune diseases, allergies, transplant rejection, or immune-mediated rejection of a therapeutics optionally in combination with an additional therapeutic agent. Furthermore, the anti-CD79 mAbs and fragments thereof can be used for diagnostic purposes, including to detect CD79 cells in biological samples and to diagnose B-cell associated disorders.

A method of inducing anergy in B-cells, comprising the steps of: providing a mammal, and administering to the mammal an anti-CD79 antibody comprising a heavy chain with a variable region selected from the group consisting of SEQ ID NOs: 9-17, 24-27, 32-41, 71, 72, and 75-77, a light chain with a variable region selected from the group consisting of SEQ ID NOs: 18-23, 28-31, 42-56, and 73-74, whereby the anti-CD79 antibody induces an anergic state in the mammal's B-cells. 
     
     
         23 . The method of  claim 22 , wherein the variable region of the heavy chain is a SEQ ID NO: 25 and the variable region of the light chain is a SEQ ID NO: 30. 
     
     
         24 . The method of  claim 22 , wherein the variable region of the heavy chain is a SEQ ID NO: 75 and the variable region of the light chain is a SEQ ID NO: 30. 
     
     
         25 . The method of  claim 22 , wherein the mammal has an arthritis, a systemic lupus, a type I diabetes, or a multiple sclerosis. 
     
     
         26 . The method of  claim 25 , wherein the variable region of the heavy chain is a SEQ ID NO: 25 and the variable region of the light chain is a SEQ ID NO: 30. 
     
     
         27 . The method of  claim 25 , wherein the variable region of the heavy chain is a SEQ ID NO: 75 and the variable region of the light chain is a SEQ ID NO: 30.
