Abstract

A cryopreserved cartilage product is disclosed. The cryopreserved cartilage product can include a partially digested cryopreserved natural cartilage collagen matrix isolated from a subject. The collagen matrix can include viable cells embedded within the collagen matrix that are native to the collagen matrix and that were embedded in the collagen matrix when the collagen matrix was isolated from the subject, at least 70% of the embedded cells native to the collagen matrix can be viable in the cryopreserved cartilage product, and the partially digested collagen matrix can retain interaction between the collagen matrix and the native cells.

Claims

exact text as granted — not AI-modified
What is claimed:

1 . A cryopreserved cartilage product comprising a partially digested cryopreserved natural cartilage collagen matrix isolated from a subject, wherein:
the collagen matrix contains viable cells embedded within the collagen matrix that are native to the collagen matrix and that were embedded in the collagen matrix when the collagen matrix was isolated from the subject; at least 70% of the embedded cells native to the collagen matrix are viable in the cryopreserved cartilage product; and the partially digested collagen matrix retains interaction between the collagen matrix and the native cells.

2 . The cryopreserved cartilage product of claim 1 , wherein the collagen matrix has a thickness of about 0.2 mm to about 2.0 mm.

3 . The cryopreserved cartilage product of claim 1 , wherein the collagen matrix has a thickness of about 1 mm to about 1.5 mm.

4 . The cryopreserved cartilage product of claim 1 , wherein the viable cells native to the collagen matrix comprise viable chondrocytes.

5 . The cryopreserved cartilage product of claim 1 , wherein the collagen matrix comprises a bioactive factor that is native to the collagen matrix.

6 . The cryopreserved cartilage product of claim 1 , further comprising a cryopreservation medium.

7 . The cryopreserved cartilage product of claim 1 , wherein the cartilage product is devoid of subchondral bone or calcified cartilage.

8 . The cryopreserved cartilage product of claim 1 , wherein the cartilage product is depleted of immunogenic cells.

9 . The cryopreserved cartilage product of claim 1 , wherein the cartilage product does not comprise macrophages.

10 . The cryopreserved cartilage product of claim 1 , further comprising a layer of bone, wherein the layer of bone is native to the collagen matrix.

11 . The cryopreserved cartilage product of claim 1 , wherein the partially digested collagen matrix is substantially intact relative to an undigested collagen matrix.

12 . The cryopreserved cartilage product of claim 1 , wherein the partially digested collagen matrix comprises extracellular matrix protein fragments.

13 . The cryopreserved cartilage product of claim 1 , wherein the collagen matrix comprises a cartilage layer that includes a radial layer, a transitional layer, or a tangential layer.

14 . The cryopreserved cartilage product of claim 13 , wherein the collagen matrix comprises a plurality of channels.

15 . The cryopreserved cartilage product of claim 14 , wherein the plurality of channels extend entirely through the radial, transitional, or tangential layer.

16 . The cryopreserved cartilage product of claim 14 , wherein the plurality of channels have a cylindrical shape.

17 . The cryopreserved cartilage product of claim 14 , wherein the collagen matrix comprises a channel density of about 10 to about 100.

18 . The cryopreserved cartilage product of claim 14 , wherein the plurality of channels have a diameter of about 0.6 mm to about 1 mm.

19 . A method of treating an injured tissue in a subject, the method comprising removing the injured tissue and administering a thawed cryopreserved cartilage product of claim 1 to the site where the injured tissue was removed.

20 . The method of claim 19 , wherein the injured tissue is articular cartilage.
