Abstract

Provided herein are improved compositions and methods for enzyme replacement therapy using modified human cystathionine beta synthase (CBS) in the treatment of homocystinuria and related diseases and disorders.

A composition comprising a carboxyl-terminal truncated cystathionine-β-synthase (CBS) polypeptide comprising the amino acid sequence SEQ ID NO: 3, wherein said truncated CBS polypeptide is covalently linked to one or a plurality of polyethylene glycol molecules, and each of the one or the plurality of polyethylene glycol has a mass of less than or equal to 5 kDa.
