Abstract
The present invention provides liposome compositions containing substituted ammonium and/or polyanion, and optionally with a desired therapeutic or imaging entity. The present invention also provide methods of making the liposome compositions provided by the present invention.

21 . A pharmaceutical composition comprising irinotecan liposomes encapsulating the irinotecan and a polyphosphate having 13-18 phosphate units per polyphosphate molecule, wherein the millimoles of irinotecan per gram of total liposomal lipids is between 0.8 and 1.7.
22 . The composition of claim 21 , comprising a lecithin and cholesterol.
23 . The composition of claim 22 , wherein the lecithin:cholesterol molar ratio is 3:2.
24 . The composition of claim 21 , comprising DSPC and cholesterol in a 3:2 molar ratio.
25 . The composition of claim 21 , comprising DSPC, cholesterol, and an amphipathic polymer comprising a polyethylene glycol-lipid.
26 . The composition of claim 25 , wherein the polyethylene glycol-lipid is a polyethylene glycol phosphatidylethanolamine.
27 . The composition of claim 26 , wherein the polyethylene glycol phosphatidylethanolamine has a poly(ethylene glycol) moiety having a molecular weight ranging from about 250 to about 20,000.
28 . The composition of claim 21 , comprising DSPC, cholesterol, and N-(methoxy-poly(ethylene glycol)-oxycarbonyl)-di stearoylphosphatidylethanolamine, in a molar ratio of 3:2:0.015.
29 . The composition of claim 21 , comprising DSPC, cholesterol, and methoxy-PEG(2000)-DSPE, in a molar ratio of 3:2:0.015.
30 . The composition of claim 21 , comprising one or more phospholipids, and wherein the liposomes encapsulate irinotecan and a polyphosphate having 13-18 phosphate units per polyphosphate molecule, and the ratio of grams of irinotecan to moles of total phospholipids is (134.4±9.3):1.
31 . The composition of claim 21 , comprising one or more phospholipids, and wherein the liposomes encapsulate irinotecan and a polyphosphate having 13-18 phosphate units per polyphosphate molecule, and the ratio of grams of irinotecan to moles of total phospholipids is (162.4±4.0):1.
