Treatment of brain cancer

Abstract

Compounds for the treatment of brain cancer are provided herein. Pharmaceutical compositions comprised of those compounds for the treatment of brain cancer are also provided herein.

         31 . A method of treating brain metastases in a patient suffering from ErbB2 positive breast cancer with brain metastases, comprising administering to the patient a therapeutically effective amount of N4-(4-([1,2,4]triazolo[1,5-a]pyridin-7-yloxy)-3-methylphenyl)-N6-(4,4-dimethyl-4,5-dihydrooxazol-2-yl)quinazoline-4,6-diamine, in combination with trastuzumab and capecitabine, wherein the patient has been previously treated with trastuzumab emtansine for breast cancer. 
     
     
         32 . The method of  claim 31 , wherein the patient has been previously treated with one or more additional therapeutic agent. 
     
     
         33 . The method of  claim 31 , wherein the one or more additional therapeutic agent is trastuzumab. 
     
     
         34 . The method of  claim 31 , wherein N4-(4-([1,2,4]triazolo[1,5-a]pyridin-7-yloxy)-3-methylphenyl)-N6-(4,4-dimethyl-4,5-dihydrooxazol-2-yl)quinazoline-4,6-diamine is administered to the patient in an oral dosage form. 
     
     
         35 . The method of  claim 34 , wherein the oral dosage form is administered from about 100 mg to about 1600 mg per day. 
     
     
         36 . The method of  claim 32 , wherein N4-(4-([1,2,4]triazolo[1,5-a]pyridin-7-yloxy)-3-methylphenyl)-N6-(4,4-dimethyl-4,5-dihydrooxazol-2-yl)quinazoline-4,6-diamine is administered to the patient in an oral dosage form. 
     
     
         37 . The method of  claim 36 , wherein the oral dosage form is administered from about 100 mg to about 1600 mg per day. 
     
     
         38 . The method of  claim 33 , wherein N4-(4-([1,2,4]triazolo[1,5-a]pyridin-7-yloxy)-3-methylphenyl)-N6-(4,4-dimethyl-4,5-dihydrooxazol-2-yl)quinazoline-4,6-diamine is administered to the patient in an oral dosage form. 
     
     
         39 . The method of  claim 38 , wherein the oral dosage form is administered from about 100 mg to about 1600 mg per day. 
     
     
         40 . A method of treating a patient suffering from breast cancer with brain metastases, comprising administering to the patient a therapeutically effective amount of N4-(4-([1,2,4]triazolo[1,5-a]pyridin-7-yloxy)-3-methylphenyl)-N6-(4,4-dimethyl-4,5-dihydrooxazol-2-yl)quinazoline-4,6-diamine, in combination with trastuzumab and capecitabine, wherein the patient has been previously treated with trastuzumab emtansine for breast cancer. 
     
     
         41 . The method of  claim 40 , wherein the breast cancer is ErbB2 positive. 
     
     
         42 . The method of  claim 41 , wherein the patient has been previously treated with one or more additional therapeutic agent. 
     
     
         43 . The method of  claim 42 , wherein the one or more additional therapeutic agent is trastuzumab. 
     
     
         44 . The method of  claim 41 , wherein N4-(4-([1,2,4]triazolo[1,5-a]pyridin-7-yloxy)-3-methylphenyl)-N6-(4,4-dimethyl-4,5-dihydrooxazol-2-yl)quinazoline-4,6-diamine is administered to the patient in an oral dosage form. 
     
     
         45 . The method of  claim 44 , wherein the oral dosage form is administered from about 100 mg to about 1600 mg per day. 
     
     
         46 . The method of  claim 42 , wherein N4-(4-([1,2,4]triazolo[1,5-a]pyridin-7-yloxy)-3-methylphenyl)-N6-(4,4-dimethyl-4,5-dihydrooxazol-2-yl)quinazoline-4,6-diamine is administered to the patient in an oral dosage form. 
     
     
         47 . The method of  claim 46 , wherein the oral dosage form is administered from about 100 mg to about 1600 mg per day. 
     
     
         48 . The method of  claim 43 , wherein N4-(4-([1,2,4]triazolo[1,5-a]pyridin-7-yloxy)-3-methylphenyl)-N6-(4,4-dimethyl-4,5-dihydrooxazol-2-yl)quinazoline-4,6-diamine is administered to the patient in an oral dosage form. 
     
     
         49 . The method of  claim 48 , wherein the oral dosage form is administered from about 100 mg to about 1600 mg per day.
