Abstract
Disclosed herein are antisense compounds and methods for decreasing Factor 11 and treating or preventing thromboembolic complications in an individual in need thereof. Examples of disease conditions that can be ameliorated with the administration of antisense compounds targeted to Factor 11 include thrombosis, embolism, and thromboembolism, such as, deep vein thrombosis, pulmonary embolism, myocardial infarction, and stroke. Antisense compounds targeting Factor 11 can also be used as a prophylactic treatment to prevent individuals at risk for thrombosis and embolism.

55 . A method of treating a thromboembolic condition in a patient, by prolonging activated partial thromboplastin time (aPTT), comprising administering to the individual an effective amount of a Factor XI inhibitor compound comprising at least one heterocyclic base moiety, wherein aPTT is prolonged by at least 20%, wherein said administering does not significantly prolong prothrombin time (PT) in the patient.
56 . The method of claim 55 , wherein aPTT is prolonged in a dose-dependent manner.
57 . The method of claim 55 , whereby bleeding risk, as determined by a bleed volume, is not increased when compared to untreated individuals.
58 . The method of claim 57 , whereby bleeding risk, as determined by a bleed volume, is increased not more than 3-fold when compared to untreated individuals.
59 . The method of claim 55 , wherein the Factor XI inhibitor compound is co-administered with a compound selected from the group consisting of aspirin, clopidogrel, dipyridamole, heparin, lepirudin, ticlopidine, warfarin, apixaban, rivaroxaban, and LOVENOX.
60 . The method of claim 55 , whereby Platelet Factor 4 (PF-4) is decreased.
