Abstract
Branching phosphoramidite monomers and molecules having comb-like structures are disclosed and described. A branching phosphoramidite monomer having the structure is provided wherein R4 and R5 are independently —(O—CH 2 —CH 2 —) n where n is 1-5 or —O—(CH 2 —) n where n is 1-10, and R1, R2, and R3 are each one of dimethoxytrityl (DMT)-O—, levulinyl (Lev)-O—, and a phosphoramidite.
Claims
exact text as granted — not AI-modifiedWhat is claimed is:
1 . A comb-like branching molecule having the structure
wherein R1 is —((CH 2 —CH 2 —) n )—R5 where n is 1-5, —((CH 2 —) n )—R5 where n is 1-10, a phosphoramidite, or a combination thereof;
R2 is —(CH 2 —CH 2 —) n where n is 1-5 or —(CH 2 —) n where n is 1-10;
R3 is a spacer;
R4 is an oligonucleotide sequence; and
R5 is a solid substrate, a reporter molecule, an oligonucleotide, or a combination thereof.
2 . The comb-like branching molecule of claim 1 , wherein the spacer (R3) is —(CH 2 —CH 2 ) n where n is 1-5, —(CH 2 —) n where n is 1-10, a phosphoramidite, an oligonucleotide, or a combination thereof.
3 . The comb-like branching molecule of claim 1 , wherein the spacer includes a cleavable linker.
4 . The comb-like branching molecule of claim 1 , wherein the oligonucleotide sequence comprises a barcode sequence.
5 . The comb-like branching molecule of claim 1 , wherein R5 is a solid substrate.Join the waitlist — get patent alerts
