Abstract

Disclosed herein are tricyclic compounds, including pyrimido[4′,5′:4,5]thieno[2,3-c]pyridazine-8-amine, pyrido[3′,2′:4,5]thieno[3,2-d]pyrimidine-4-amine, pyrazino[2′,3′:4,5]thieno[3,2-d]pyrimidin-4-amine, pyrido[3′,2′:4,5]furo[3,2-d]pyrimidin-4-amine, and pyrimido[4′,5′:4,5]furo[2,3-c]pyridazin-8-amine compounds, which may be useful as positive allosteric modulators of the muscarinic acetylcholine receptor M4 (mAChR M4). Also disclosed herein are methods of making the compounds, pharmaceutical compositions comprising the compounds, and methods of treating neurological and psychiatric disorders associated with muscarinic acetylcholine receptor dysfunction using the compounds and compositions.

What is claimed is: 
     
         1 . A compound of formula (I) 
       
         
           
           
               
               
           
         
         or a pharmaceutically acceptable salt thereof, wherein: 
         B is O or S; 
         X is N or CR 1 ; 
         Z is N or CR 3 ; 
         R 1  is selected from hydrogen, C 1 -C 4 -alkyl, C 1 -C 4 -haloalkyl, halo, and —OR b ; 
         R 2  is selected from hydrogen, C 1 -C 4 -alkyl, C 1 -C 4 -haloalkyl, C 1 -C 4 -hydroxyalkyl, C 1 -C 4 -acetoxyalkyl, halo, and —OR b ; 
         R 3  is selected from hydrogen and C 1 -C 4 -alkyl; 
         R 4  and R 5  are each independently selected from hydrogen, alkyl, alkenyl, alkynyl, aryl, heteroaryl, cycloalkyl, heteroalkyl, heterocycle, and —(CR c R d ) n —Y, each of which may be optionally substituted; 
         or R 4  and R 5  are taken together with the nitrogen atom to which they are attached to form an optionally substituted heterocyclic ring; 
         R 6  is selected from hydrogen and C 1 -C 4 -alkyl; 
         each Y is independently selected from halo, —OR, —SR, —C(O)R, —C(O)OR, —S(O)R, —SO 2 R, —NR 2 , —C(O)NR 2 , —NRC(O)R, —S(O) 2 NR 2 , —NRS(O) 2 R, aryl, heteroaryl, cycloalkyl, and heterocycle, each of which may be optionally substituted; 
         n is 1, 2, 3, 4, 5, 6, 7, or 8; 
         R b  is selected from hydrogen, C 1 -C 4 -alkyl, C 1 -C 4 -haloalkyl, aryl, heteroaryl, cycloalkyl, and heterocycle; 
         R c  and R d  are each independently selected from hydrogen, C 1 -C 4 -alkyl, C 1 -C 4 -haloalkyl, and halo; and 
         each R is independently selected from hydrogen, alkyl, aryl, arylalkyl, cycloalkyl, cycloalkylalkyl, heterocycle, heterocyclealkyl, heteroaryl, heteroarylalkyl, and heteroalkyl, each of which may be optionally substituted.

         25 . The compound of  claim 24 , wherein
 R 4  is selected from hydrogen and methyl;   R 5  is selected from C 1 -C 4  alkyl, C 3 -C 6  cycloalkyl, a 3- to 6-membered heterocycle having one heteroatom selected from N and O, and —(CR c R d ) n —Y;   n is 1, 2 or 3;   R c  and R d  are each hydrogen;   Y is selected from —OR, aryl, C 3 -C 6  cycloalkyl, a 5- to 6-membered heteroaryl having 1 or 2 heteroatoms independently selected from N, O and S, a 3- to 6-membered heterocycle having one or two heteroatoms independently selected from N, O and S, sulfanyl, —COR, —C(O)NR 2 , and —SO 2 R;   wherein each alkyl, aryl, cycloalkyl, heteroaryl and heterocycle is independently unsubstituted or substituted with 1 or 2 substituents independently selected from C 1 -C 4  alkyl, C 1 -C 4  hydroxyalkyl, C 1 -C 4  alkoxy, C 1 -C 4 -alkoxy-C 1 -C 4 -alkyl, C 1 -C 4 -aminoalkyl, —CONR 2 , halo, hydroxy, aryl, C 3 -C 6  cycloalkyl, a 3- to 6-membered heterocycle having one heteroatom independently selected from N and O, and a 5- to 6-membered heteroaryl having 1 or 2 heteroatoms independently selected from N, O and S; and   each R is independently selected from hydrogen, C 1 -C 4  alkyl, C 1 -C 4  hydroxyalkyl, C 1 -C 4  haloalkyl, C 1 -C 4 -alkoxy-C 1 -C 4 -alkyl, C 3 -C 6  cycloalkyl, a 5- to 6-membered heteroaryl group having 1, 2 or 3 heteroatoms independently selected from N, S and O, a 3- to 6-membered heterocycle having one or two heteroatoms independently selected from N and O, or a bicyclic heterocycle, C 3 -C 6  cycloalkyl.   
     
     
         26 . The compound of  claim 24 , wherein
 R 4  is selected from hydrogen and methyl;   R 5  is selected from C 1 -C 4  alkyl, C 3 -C 6  cycloalkyl, a 3- to 6-membered heterocycle having one heteroatom selected from N and O, and —(CR c R d ) n —Y;   n is 1, 2 or 3;   R c  and R d  are each hydrogen;   Y is selected from —OR, aryl, C 3 -C 6  cycloalkyl, and a 5- to 6-membered heteroaryl having 1 or 2 heteroatoms independently selected from N, O and S; and   R is C 1 -C 4  alkyl;   wherein each aryl, cycloalkyl, heteroaryl and heterocycle is independently unsubstituted or substituted with 1 or 2 substituents independently selected from C 1 -C 4  alkyl, C 1 -C 4  hydroxyalkyl, C 1 -C 4  alkoxy, halo, hydroxy, aryl, C 3 -C 6  cycloalkyl, a 3- to 6-membered heterocycle having one heteroatom independently selected from N and O, and a 5- to 6-membered heteroaryl having 1 or 2 heteroatoms independently selected from N, O and S.   
     
     
         27 . The compound of any one of  claims 1 - 23 , wherein
 R 4  and R 5  are taken together with the nitrogen atom to which they are attached to form an azetidinyl, pyrrolidinyl, piperidinyl or piperazinyl group, which may be optionally substituted with 1, 2 or 3 substituents independently selected from alkyl, alkenyl, alkynyl, aryl, heteroaryl, cycloalkyl, heteroalkyl, heterocycle, oxo, and —(CR c R d ) p —Y, wherein p is 0, 1, 2, 3, 4, 5, 6, 7, or 8.   
     
     
         28 . The compound of  claim 27 , wherein
 R 4  and R 5  are taken together with the nitrogen atom to which they are attached to form an azetidinyl, pyrrolidinyl, piperidinyl or piperazinyl group, each of which is independently unsubstituted or substituted with 1 or 2 substituents independently selected from C 1 -C 4  alkyl, C 3 -C 6  cycloalkyl, aryl, a 5- to 6-membered heteroaryl having 1 or 2 heteroatoms independently selected from N, O and S, a 3- to 6-membered heterocycle having one heteroatom independently selected from N and O, oxo, and —(CR c R d ) p —Y;   p is 0 or 1;   R c  and R d  are each hydrogen;   each Y is independently selected from C 1 -C 4  alkyl, aryl, —OR, —NR 2 ; and   each R is independently selected from hydrogen, C 1 -C 4  alkyl, aryl, and a 5- to 6-membered heteroaryl having 1 or 2 heteroatoms independently selected from N, O and S;   wherein each aryl, cycloalkyl, heteroaryl and heterocycle is independently unsubstituted or substituted with 1 or 2 substituents independently selected from C 1 -C 4  alkyl, C 1 -C 4  haloalkyl, cyano, and halo.   
     
     
         29 . The compound of any one of  claims 1 - 28 , wherein
 R 6  is hydrogen.

 A is selected from phenyl, a C 3 -C 6  cycloalkyl, a 3- to 6-membered heterocycle having one or two heteroatoms independently selected from N, S and O, and a 5- to 6-membered heteroaryl having 1 or 2 heteroatoms independently selected from N, O and S.   
     
     
         40 . The compound of any one of  claims 38 - 39 , wherein

A method for treating a neurological and/or psychiatric disorder associated with muscarinic acetylcholine receptor dysfunction in a mammal, comprising a step of administering to the mammal a therapeutically effective amount a compound of any one of  claims 1 - 48 , or pharmaceutically acceptable salt thereof. 
     
     
         51 . The method of  claim 50 , wherein the disorder is associated with a mAChR M 4  dysfunction. 
     
     
         52 . The method of  claim 50 , wherein the disorder is a neurological and/or psychiatric disorder associated with mAChR M 4  dysfunction. 
     
     
         53 . The method of  claim 50 , wherein the disorder is selected from Alzheimer's disease, schizophrenia, a sleep disorder, a pain disorder, and a cognitive disorder. 
     
     
         54 . The method of  claim 53 , wherein the disorder is Alzheimer's disease.

A kit comprising a compound of any one of  claims 1 - 48 , or pharmaceutically acceptable salt thereof, and one or more of: (a) at least one agent known to increase mAChR M 4  activity; (b) at least one agent known to decrease mAChR M 4  activity; (c) at least one agent known to treat a disorder associated with cholinergic activity; (d) instructions for treating a disorder associated with cholinergic activity; (e) instructions for treating a disorder associated with mAChR M 4  receptor activity; and (f) instructions for administering the compound in connection with cognitive or behavioral therapy.
