Abstract

The present disclosure generally relates to the use of different self-amplifying RNA molecules to enhance immune responses, for example immune responses following prophylactic vaccination or therapeutic administration. Some embodiments relate to compositions and methods for inducing an immune response in a subject using prime-boost immunization regimens.

What is claimed is: 
     
         1 . A composition comprising:
 a priming composition comprising a first RNA replicon which encodes a first antigen; and   a boosting composition comprising a second RNA replicon which encodes a second   antigen wherein the first and second RNA replicons are different from each other.   
     
     
         2 . A composition comprising:
 a first nucleic acid sequence encoding a first RNA replicon which encodes a first antigen; and   a second nucleic acid sequence encoding a second RNA replicon which encodes a second antigen,   wherein the first and second RNA replicons are different from each other,   wherein the first replicon and/or the second replicon comprises at least one expression cassettes comprising a promoter operably linked to a coding sequence for a molecule of interest.
