Claims

Abstract

N2-naphthalenesulfonyl-L-arginine esters and amides having the formula <IMAGE> 1 or the acid addition salts thereof with a pharmaceutically acceptable acid, wherein R is selected from the class consisting of (1) alkoxy, alkenyloxy, cycloalkoxy and halogenated alkoxy, respectively containing not more than 10 carbon atoms, aralkyloxy of not more than 15 carbon atoms, and alkoxy of not more than 10 carbon atoms substitured with an alkoxy group of not more than 10 carbon atoms; <IMAGE> (2) wherein R1 and R2 are members selected from the class consisting of hydrogen, alkyl, aryl, alkenyl and cycloalkyl, respectively containing not more than 10 carbon atoms, and aralkyl and cycloalkylalkyl, respectively containing not more than 15 carbon atoms, and substituted alkyl containing not more than 20 carbon atoms, said substituent being selected from the class consisting of alkoxy, alkoxycarbonyl, acyl, acyloxy, arylcarbamoyl and N,N-polymethylenecarbamoyl, respectively containing not more than 10 carbon atoms, and carboxy; and <IMAGE> (3) wherein Z is a divalent group containing up to 10 carbons atoms, which consists of more than one group selected from the class consisting of methylene -CH2-, monosubstituted methylene <IMAGE> wherein R3 is selected from the class consisting of alkyl, acyl, alkoxy, and alkoxycarbonyl, respectively containing not more than 10 carbon atoms, and carbamoyl; and disubstituted methylene <IMAGE> wherein R4 and R5 are alkyl groups of not more than 10 carbon atoms, and which may further contain at least one member selected from the class consisting of oxy-O-, thio -S-, cycloalkylene of not more than 10 carbon atoms, imino <IMAGE> alkyl-substituted imino <IMAGE> wherein R6 is an alkyl group of not more than 10 carbon atoms, acyl-substituted imino <IMAGE> wherein R7 is an alkyl group of not more than 10 carbon atoms, and phenylene <IMAGE> which may be arranged in any order and complete the <IMAGE> ring together with the said methylene, monosubstituted methylene or disubstituted methylene; and R' is a member selected from the class consisting of 1-naphthyl substituted alkoxy or alkoxycarbonyloxy, respectively containing not more than 10 carbon atoms; and 2-naphthyl substituted with alkoxy or alkoxycarbonyloxy, respectively containing not more than 10 carbon atoms.

what is claimed as new and intended to be covered by letters patent is: 
     
       1. N 2  -naphthalenesulfonyl-L-arginine amides having the formula ##STR67## or the acid addition salts thereof with a pharmaceutically acceptable acid, wherein R 1  and R 2  are members selected from the group consisting of hydrogen and a hydrocarbon selected from the group consisting of C 1-10  alkyl, C 6-10  aryl, C 7-15  aralkyl, C 3-10  cycloalkyl and C 4-15  cycloalkylalkyl, or R 1  or R 2  together form a polymethyleneiminyl group of 3-10 carbon atoms, the methylene groups of which are optionally substituted by one or two alkyl groups of not more than 10 carbon atoms; and R' is a member selected from the class consisting of 1-naphthyl substituted with one member selected from the class consisting of alkoxy and alkoxycarbonyloxy, respectively containing not more than 10 carbon atoms; and 2-naphthyl substituted with a member selected from the class consisting of alkoxy and alkoxycarbonyloxy, respectively containing not more than 10 carbon atoms. 
     
     
       2. The compounds of claim 1, wherein R 1  is hydrogen and R 2  is a member selected from the class consisting of C 1-10  alkyl, C 6-10  aryl, C 7-15  aralkyl, C 3-10  cycloalkyl and C 4-15  cycloalkylalkyl. 
     
     
       3. The compounds of claim 2, wherein R 2  is a member selected from the class consisting of C 1-9  alkyl, C 3-6  cycloalkyl, C 6-10  aryl, C 7-10  aralkyl, C 7-10  cycloalkylalkyl and R' is a member selected from the class consisting of 1-naphthyl substituted with a member selected from the class consisting of alkoxy and alkoxycarbonyloxy, respectively containing not more than 5 carbon atoms and 2-naphthyl substituted with a member selected from the class consisting of alkoxy and alkoxycarbonyloxy, respectively containing not more than 5 carbon atoms. 
     
     
       4. The compounds of claim 3, wherein R 2  is a member selected from the class consisting of butyl, cyclohexylmethyl and benzyl and R 1  is a member selected from the class consisting of 4-methoxy-1-naphthyl, 4-methoxy-2-naphthyl, 5-methoxy-1-naphthyl, 6-methoxy-2-naphthyl, 7-methoxy-2-naphthyl, 1-methoxy-2-naphthyl, 5-ethoxy-1-naphthyl, 5-propoxy-1-naphthyl, 5-isopropoxy-1-naphthyl, 5-ethoxycarbonyloxy-1-naphthyl and 5-ethoxycarbonyloxy-2-naphthyl. 
     
     
       5. The compounds of claim 1, wherein R 1  and R 2  are respectively a member selected from the class consisting of C 1-10  alkyl, C 6-10  aryl, C 7-15  aralkyl and C 4-15  cycloalkylalkyl. 
     
     
       6. The compounds of claim 5, wherein the entity ##STR68## in the formula is a member selected from the class consisting of dialkylamino of 2-10 carbon atoms, and N-alkyl-N-aralkylamino of 8-10 carbon atoms and R' is a group selected from the class consisting of 1-naphthyl substituted with one member selected from the class consisting of alkoxy and alkoxycarbonyloxy, respectively containing not more than 5 carbon atoms, and 2-naphthyl substituted with one member selected from the class consisting of alkoxy and alkoxycarbonyloxy, respectively containing not more than 5 carbon atoms. 
     
     
       7. The compounds of claim 6, wherein the entity ##STR69## in the formula is a member selected from the class consisting of N-methyl-N-butylamino, and N-methyl-N-benzylamino; and R' is a member selected from the class consisting of 4-methoxy-1-naphthyl, 4-methoxy-2-naphthyl, 5-methoxy-1-naphthyl, 6-methoxy-2-naphthyl, 7-methoxy-2-naphthyl, 1-methoxy-2-naphthyl, 5-ethoxy-1-naphthyl, 5-propoxy-1-naphthyl, 5-isopropoxy-1-naphthyl, 5-ethoxycarbonyloxy-1-naphthyl and 5-ethoxycarbonyloxy-2-naphthyl. 
     
     
       8. The compounds of claim 1, wherein R 1  and R 2  together form a polymethyleneiminyl group of 3-10 carbon atoms, the methylene groups of which are optionally substituted by one or two alkyl groups of not more than 10 carbon atoms. 
     
     
       9. The compound of claim 8 wherein ##STR70## is a member selected from the class consisting of N,N-polymethyleniminyl of 3-10 carbon atoms and N,N-polymethyleniminyl of 3-10 carbon atoms whose methylene groups are substituted with alkyl of 1-5 carbon atoms; and R' is a group selected from the class consisting of 1-naphthyl substituted with one member selected from the class consisting of alkoxy and alkoxycarbonyloxy, respectively containing not more than 5 carbon atoms and 2-naphthyl substituted with one member selected from the class consisting of alkoxy and alkoxycarbonyloxy, respectively containing not more than 5 carbon atoms. 
     
     
       10. The compounds of claim 9, wherein ##STR71## is a member selected from the class consisting of piperidino, hexamethyleniminyl, heptamethyleniminyl, octamethyleniminyl, 4-methylpiperidino, 4-ethylpiperidino, and 4-ethylpiperidino, and R' is a member selected from the class consisting of 4-methoxy-1-naphthyl, 4-methoxy-2-naphthyl, 5-methoxy-1-naphthyl, 6-methoxy-2-naphthyl, 7-methoxy-2-naphthyl, 1-methoxy-2-naphthyl, 5-ethoxy-1-naphthyl, 5-propoxy-1-naphthyl, 5-isopropoxy-1-naphthyl, 5-ethoxycarbonyloxy-1-naphthyl and 5-ethoxycarbonyloxy-2-naphthyl. 
     
     
       11. A method for inhibiting activity and suppressing of activation of thrombin in vivo, comprising administering to a patient a pharmaceutically effective amount of a compound of claim 1.
