Abstract
The present invention relates to a novel human protein called Dendritic Enriched Secreted Lymphocyte Activation Molecule, and isolated polynucleotides encoding this protein. Also provided are vectors, host cells, antibodies, and recombinant methods for producing this human protein. The invention further relates to diagnostic and therapeutic methods useful for diagnosing and treating disorders related to this novel human protein.

exact text as granted — not AI-modified1. An isolated antibody or fragment thereof that specifically binds to a protein selected from the group consisting of:
(a) a protein consisting of amino acid residues 1 to 285 of SEQ ID NO:2;
(b) a protein consisting of amino acid residues 23 to 285 of SEQ ID NO:2;
(c) a protein consisting of amino acid residues 23 to 232 of SEQ ID NO:2;
(d) a protein consisting of a portion of SEQ ID NO:2, wherein said portion is at least 30 contiguous amino acid residues of SEQ ID NO:2 in length; and
(e) a protein consisting of a portion of SEQ ID NO:2, wherein said portion is at least 50 contiguous amino acid residues of SEQ ID NO:2 in length.
2. The antibody or fragment thereof of claim 1 that specifically binds protein (a).
3. The antibody or fragment thereof of claim 1 that specifically binds protein (b).
4. The antibody or fragment thereof of claim 1 that specifically binds protein (c).
5. The antibody or fragment thereof of claim 1 that specifically binds protein (d).
6. The antibody or fragment thereof of claim 1 that specifically binds protein (e).
7. The antibody or fragment thereof of claim 2 that specifically binds protein (b).
8. The antibody or fragment thereof of claim 4 wherein said protein bound by said antibody or fragment thereof is glycosylated.
9. The antibody or fragment thereof of claim 4 which is a human antibody.
10. The antibody or fragment thereof of claim 4 which is a polyclonal antibody.
11. The antibody or fragment thereof of claim 4 which is selected from the group consisting of:
(a) a chimeric antibody;
(b) a humanized antibody;
(c) a single chain antibody; and
(d) a Fab fragment.
12. The antibody or fragment thereof of claim 4 which is coupled or conjugated to a detectable substance.
13. The antibody of fragment thereof of claim 12 wherein the detectable substance is selected from the group consisting of:
(a) an enzyme;
(b) a fluorescent label;
(c) a luminescent label;
(d) a bioluminescent label;
(e) a radioactive label; and
(f) a prosthetic label.
14. The antibody or fragment thereof of claim 4 wherein said antibody or fragment thereof specifically binds to said protein in a Western blot.
15. The antibody or fragment thereof of claim 4 wherein said antibody or fragment thereof specifically binds to said protein in an ELISA.
16. An isolated cell that produces the antibody or fragment thereof of claim 4 .
17. A hybridoma that produces the antibody or fragment thereof of claim 4 .
18. A method of detecting a protein in a biological sample comprising:
(a) contacting the biological sample with the antibody or fragment thereof of claim 4 ; and
(b) detecting the protein in the biological sample.
19. The method of claim 18 wherein the antibody or fragment thereof is a polyclonal antibody.
20. An isolated antibody or fragment thereof obtained from an animal that has been immunized with a protein selected from the group consisting of:
(a) a protein consisting of amino acid residues 1 to 285 of SEQ ID NO:2;
(b) a protein consisting of amino acid residues 23 to 285 of SEQ ID NO:2;
(c) a protein consisting of amino acid residues 23 to 232 of SEQ ID NO:2;
(d) a protein consisting of a portion of SEQ ID NO:2, wherein said portion is at least 30 contiguous amino acid residues of SEQ ID NO:2 in length; and
(e) a protein consisting of a portion of SEQ ID NO:2, wherein said portion is at least 50 contiguous amino acid residues of SEQ ID NO:2 in length;
wherein said antibody or fragment thereof specifically binds to said amino acid sequence.
21. The antibody or fragment thereof of claim 20 obtained from an animal immunized with protein (a).
22. The antibody or fragment thereof of claim 20 obtained from an animal immunized with protein (b).
23. The antibody or fragment thereof of claim 20 obtained from an animal immunized with protein (c).
24. The antibody or fragment thereof of claim 20 obtained from an animal immunized with protein (d).

28. An isolated monoclonal antibody or fragment thereof that specifically binds to a protein selected from the group consisting of:
(a) a protein consisting of amino acid residues 1 to 285 of SEQ ID NO:2;
(b) a protein consisting of amino acid residues 23 to 285 of SEQ ID NO:2;
(c) a protein consisting of amino acid residues 23 to 232 of SEQ ID NO:2;
(d) a protein consisting of a portion of SEQ ID NO:2, wherein said portion is at least 30 contiguous amino acid residues of SEQ ID NO:2 in length; and
(e) a protein consisting of a portion of SEQ ID NO:2, wherein said portion is at least 50 contiguous amino acid residues of SEQ ID NO:2 in length.

(a) a protein consisting of the full-length polypeptide encoded by the cDNA contained in ATCC Deposit Number 209623;
(b) a protein consisting of a portion of the polypeptide encoded by the cDNA contained in ATCC Deposit Number 209623, wherein said portion is at least 30 contiguous amino acid residues in length of the polypeptide encoded by the cDNA contained in ATCC Deposit Number 209623; and
(c) a protein consisting of a portion of the polypeptide encoded by the cDNA contained in ATCC Deposit Number 209623, wherein said portion is at least 50 contiguous amino acid residues in length of the polypeptide encoded by the cDNA contained in ATCC Deposit Number 209623.

(b) a protein consisting of a portion of the polypeptide encoded by the cDNA contained in ATCC Deposit Number 209623, wherein said portion is at least 30 contiguous amino acid residues in length of the polypeptide encoded by the cDNA contained in ATCC Deposit Number 209623; and
(c) a protein consisting of a portion of the polypeptide encoded by the cDNA contained in ATCC Deposit Number 209623, wherein said portion is at least 50 contiguous amino acid residues in length of the polypeptide encoded by the cDNA contained in ATCC Deposit Number 209623;
wherein said antibody or fragment thereof specifically binds to said amino acid sequence.

(b) a protein consisting of a portion of the polypeptide encoded by the cDNA contained in ATCC Deposit Number 209623, wherein said portion is at least 30 contiguous amino acid residues in length of the polypeptide encoded by the cDNA contained in ATCC Deposit Number 209623; and
(c) a protein consisting of a portion of the polypeptide encoded by the cDNA contained in ATCC Deposit Number 209623, wherein said portion is at least 50 contiguous amino acid residues in length of the polypeptide encoded by the cDNA contained in ATCC Deposit Number 209623.

(b) detecting the protein in the biological sample.

An isolated antibody or fragment thereof that specifically binds Dendritic Enriched Secreted Lymphocyte Activation Molecule protein purified from a cell culture wherein the cells in said cell culture comprise a polynucleotide encoding amino acids 23 to 232 of SEQ ID NO:2 operably associated with a regulatory sequence that controls the expression of said polynucleotide.
82. The antibody or fragment thereof of claim 81 which is a monoclonal antibody.
83. The antibody or fragment thereof of claim 81 which is a human antibody.
84. The antibody or fragment thereof of claim 81 which is selected from the group consisting of:
(a) a chimeric antibody;
(b) a polyclonal antibody;
(c) a humanized antibody;
(d) a single chain antibody; and

85. The antibody or fragment thereof of claim 81 wherein said antibody or fragment thereof binds to said protein in a Western blot.

87. An isolated antibody or fragment thereof that binds a protein comprising amino acid residues 23 to 232 of SEQ ID NO:2 with a dissociation constant (KD) less than or equal to 10 −8 M.
88. The isolated antibody or fragment thereof of claim 87 , wherein the isolated antibody or fragment has a dissociation constant (KD) less than or equal to 10 −9 M.
89. The isolated antibody or fragment thereof of claim 87 , wherein the isolated antibody or fragment has a dissociation constant (KD) less than or equal to 10 −10 M.
90. The isolated antibody or fragment thereof of claim 87 , wherein the isolated antibody or fragment has a dissociation constant (KD) less than or equal to 10 −11 M.
