Claims

Abstract

The invention provides an anti-human IL-23R antibody having excellent activity and/or cross-reactivity compared to conventional IL-23R antibodies and methods of using the same for treating or preventing various diseases in which human IL-23R is involved in the pathogenesis of the diseases.

The invention claimed is: 
     
       1. An anti-human IL-23R antibody or an anti-human IL-23R antibody fragment, comprising a heavy-chain variable region consisting of the amino acid sequence shown by SEQ ID NO: 5 and a light-chain variable region consisting of the amino acid sequence shown by SEQ ID NO:7. 
     
     
       2. The anti-human IL-23R antibody of  claim 1 , comprising a heavy chain consisting of the amino acid sequence shown by SEQ ID NO:33 and a light chain consisting of the amino acid sequence shown by SEQ ID NO:35. 
     
     
       3. A method for producing an anti-human IL-23R antibody or an anti-human IL-23R antibody fragment, the method comprising a step of culturing a host cell and expressing the anti-human IL-23R antibody or the anti-human IL-23R antibody fragment, wherein the host cell is selected from the group consisting of the following (a) and (b):
 (a) a host cell transformed with an expression vector comprising a polynucleotide comprising a sequence that encodes a heavy-chain variable region consisting of the amino acid sequence shown by SEQ ID NO:5 and a polynucleotide comprising a sequence that encodes a light-chain variable region consisting of the amino acid sequence shown by SEQ ID NO:7; and 
 (b) a host cell transformed with an expression vector comprising a polynucleotide comprising a sequence that encodes a heavy-chain variable region consisting of the amino acid sequence shown by SEQ ID NO:5 and an expression vector comprising a polynucleotide comprising a sequence that encodes a light-chain variable region consisting of the amino acid sequence shown by SEQ ID NO:7. 
 
     
     
       4. An anti-human IL-23R antibody or an anti-human IL-23R antibody fragment produced by the method of  claim 3 . 
     
     
       5. A pharmaceutical composition comprising an anti-IL-23R antibody or an anti-human IL-23R antibody fragment produced by a method comprising a step of culturing a host cell and expressing the anti-human IL-23R antibody or the anti-human IL-23R antibody fragment, wherein the host cell is selected from the group consisting of the following (a) and (b):
 (a) a host cell transformed with an expression vector comprising a polynucleotide comprising a sequence that encodes the heavy-chain variable region consisting of the amino acid sequence shown by SEQ ID NO: 5 and a polynucleotide comprising a sequence that encodes the light-chain variable region consisting of the amino acid sequence shown by SEQ ID NO:7; and 
 (b) a host cell transformed with an expression vector comprising a polynucleotide comprising a sequence that encodes the heavy-chain variable region consisting of the amino acid sequence shown by SEQ ID NO:5 and an expression vector comprising a polynucleotide comprising a sequence that encodes the light-chain variable region consisting of the amino acid sequence shown by SEQ ID NO:7. 
 
     
     
       6. A method for producing an anti-human IL-23R antibody, the method comprising a step of culturing a host cell and expressing the anti-human IL-23R antibody, wherein the host cell is selected from the group consisting of the following (a) and (b):
 (a) a host cell transformed with an expression vector comprising a polynucleotide comprising a sequence that encodes a heavy chain consisting of the amino acid sequence shown by SEQ ID NO:33 and a polynucleotide comprising a sequence that encodes a light chain consisting of the amino acid sequence shown by SEQ ID NO:35; and 
 (b) a host cell transformed with an expression vector comprising a polynucleotide comprising a sequence that encodes a heavy chain consisting of the amino acid sequence shown by SEQ ID NO:33 and an expression vector comprising a polynucleotide comprising a sequence that encodes a light chain consisting of the amino acid sequence shown by SEQ ID NO:35. 
 
     
     
       7. An anti-human IL-23R antibody produced by the method of  claim 6 . 
     
     
       8. A pharmaceutical composition comprising an anti-IL-23R antibody produced by a method comprising a step of culturing a host cell and expressing the anti-human IL-23R antibody, wherein the host cell is selected from the group consisting of the following (a) and (b):
 (a) a host cell transformed with an expression vector comprising a polynucleotide comprising a sequence that encodes the heavy-chain consisting of the amino acid sequence shown by SEQ ID NO:33 and a polynucleotide comprising a sequence that encodes the light-chain consisting of the amino acid sequence shown by SEQ ID NO:35; and 
 (b) a host cell transformed with an expression vector comprising a polynucleotide comprising a sequence that encodes the heavy-chain consisting of the amino acid sequence shown by SEQ ID NO:33 and an expression vector comprising a polynucleotide comprising a sequence that encodes the light-chain consisting of the amino acid sequence shown by SEQ ID NO:35. 
 
     
     
       9. A polynucleotide comprising a sequence encoding a heavy-chain variable region of an anti-human IL-23R antibody or an anti-human IL-23R antibody fragment, the heavy-chain variable region consisting of the amino acid sequence shown by SEQ ID NO:5. 
     
     
       10. An expression vector comprising the polynucleotide of  claim 9 . 
     
     
       11. A polynucleotide comprising a sequence encoding a light-chain variable region of an anti-human IL-23R antibody or an anti-human IL-23R antibody fragment, the light-chain variable region consisting of the amino acid sequence shown by SEQ ID NO:7. 
     
     
       12. An expression vector comprising the polynucleotide of  claim 11 .
