Abstract

Provided herein are novel compounds that inhibit ribonucleotide reductase (RR) by binding to RRM2 and interfering with the activity of the RRM1/RRM2 holoenzyme, as well as methods of synthesizing these novel compounds. The compounds may be used to inhibit RR activity and to treat various conditions associated with RRM2 expression, such as for example certain cancer types, mitochondrial diseases, or degenerative diseases.

What is claimed is: 
     
       1. A method of synthesizing a compound having the structure: 
       
         
           
           
               
               
           
         
       
       comprising the steps:
 (a) converting veratrole to 1-(3,4-dimethoxyphenyl)-2-phenylethanone; 
 (b) converting 1-(3,4-dimethoxyphenyl)-2-phenylethanone to 4-(3,4-dimethoxyphenyl)-5-phenylthiazol-2-amine; 
 (c) converting 4-(3,4-dimethoxyphenyl)-5-phenylthiazol-2-amine to N-(4-(3,4-dimethoxyphenyl)-5-phenylthiazol-2-yl)-3,4-dimethoxybenzamide; and 
 (d) converting N-(4-(3,4-dimethoxyphenyl)-5-phenylthiazol-2-yl)-3,4-dimethoxybenzamide to 
 
       
         
           
           
               
               
           
         
       
     
     
       2. The method of  claim 1 , wherein:
 step (a) comprises mixing veratrole with anhydrous AlCl 3 , dichloromethane, and phenylacetyl chloride to produce 1-(3,4-dimethoxyphenyl)-2-phenylethanone; 
 step (b) comprises dissolving said 1-(3,4-dimethoxyphenyl)-2-phenylethanone and pyridinium tribromide in dichloromethane, washing and drying the resultant reaction mixture, and mixing said reaction mixture with thiourea in ethanol to produce 4-(3,4-dimethoxyphenyl)-5-phenylthiazol-2-amine; 
 step (c) comprises adding said 4-(3,4-dimethoxyphenyl)-5-phenylthiazol-2-amine 4-dimethylamino pyridine and Et 3 N to an acid chloride solution comprising 3,4-dimethoxybenzoic acid and dimethylformamide treated with thionyl chloride to produce N-(4-(3,4-dimethoxyphenyl)-5-phenylthiazol-2-yl)-3,4-dimethoxybenzamide; and 
 step (d) comprises mixing N-(4-(3,4-dimethoxyphenyl)-5-phenylthiazol-2-yl)-3,4-dimethoxybenzamide with boron tribromide in toluene to produce:
