Claims

Abstract

The present invention relates to the field of Streptococcus. More specifically, the present invention relates to the identification of polypeptides and polynucleotide sequences encoding the same which are involved in the pathogenic mechanism of S. suis. The present invention also relates to the use of such polypeptides in compositions and methods for the prevention, the treatment and diagnosis of S. suis-associated diseases and infections caused by S. suis.

       1. An isolated polypeptide comprising an amino acid sequence having at least 75% identity to the amino acid sequence set forth in SEQ ID NO: 1 or in SEQ ID NO: 4.

       3. The isolated polypeptide of  claim 1 , comprising the amino acid sequence as set forth in SEQ ID NO 10. 
     
     
       4. The isolated polypeptide of  claim 1 , comprising an amino acid having at least 85% identity to the amino acid sequence set forth in SEQ ID NO 1 or in SEQ ID NO 4. 
     
     
       5. The isolated polypeptide of  claim 1 , comprising an amino acid sequence having at least 95% identity to the amino acid sequence set forth in SEQ ID NO 1. 
     
     
       6. The isolated polypeptide of  claim 1 , comprising an amino acid sequence having at least 95% identity to the amino acid sequence set forth in SEQ ID NO 4. 
     
     
       7. The isolated polypeptide of  claim 1 , wherein it elicits a protective response to a Streptococcus suis strain challenge when administered to an animal. 
     
     
       8. The isolated polypeptide of  claim 3 , comprising at least one repetitive amino acid sequence consisting of the amino acid sequence Xaa 1  Ser Xaa 3  Xaa 4  Xaa 5  Met Xaa 7  Xaa 8  Xaa 9  Xaa 10  X 11  Xaa 12  Xaa 13  Xaa 14  Xaa 15  Xaa 16  Pro Xaa 18  Xaa 19  Gln Met Xaa 22  Asn Lys Glu Xaa 26  Xaa 27  Xaa 28  Xaa 29  Xaa 30  (SEQ ID NO 9) wherein

       13. The isolated polypeptide of  claim 1 , comprising an to amino acid sequence having 100% identity to the amino acid sequence set forth in SEQ ID NO 4. 
     
     
       14. An isolated polypeptide, comprising an amino acid sequence having at least 95% identity to the amino acid sequence set forth in SEQ ID NO 2. 
     
     
       15. The isolated polypeptide of  claim 14 , comprising an amino acid sequence having 100% identity to the amino acid sequence set forth in SEQ ID NO 2. 
     
     
       16. An isolated polypeptide, comprising an amino acid sequence having at least 95% identity to the amino acid zo sequence set forth in SEQ ID NO 3. 
     
     
       17. The isolated polypeptide of  claim 16 , comprising an amino acid sequence having 100% identity to the amino acid sequence set forth in SEQ ID NO 3. 
     
     
       18. A composition comprising an acceptable carrier and at least one of the following elements:
 a polypeptide as defined in any one of  claims 1 ,  2 ,  3 ,  4 ,  5 ,  6 ,  7 , and  8  to  17 .   
     
     
       19. A composition for eliciting an immune response to Streptococcus or treating Streptococcus suis-associated diseases or infection caused by S. suis, comprising an acceptable carrier and at least one of the following elements:
 a polypeptide as defined in any one of  claims 1 ,  2 ,  3 ,  4 ,  5 ,  6 ,  7 , and  8  to  17 .   
     
     
       20. The composition of  claim 19 , further comprising an adjuvant. 
     
     
       21. A vaccine comprising a polypeptide having at least 15 contiguous amino acid residues of the amino acid sequence of SEQ ID NO: 4, and an adjuvant. 
     
     
       22. The vaccine of claim 21, wherein the polypeptide has at least 25 contiguous amino acid residues of the amino acid sequence of SEQ ID NO: 4. 
     
     
       23. The vaccine of claim 21, wherein the polypeptide has at least 35 contiguous amino acid residues of the amino acid sequence of SEQ ID NO: 4. 
     
     
       24. The vaccine of claim 21, wherein the polypeptide further includes at the C-terminal end thereof the amino acid sequence of SEQ ID NO: 9. 
     
     
       25. The vaccine of claim 24, wherein the amino acid sequence of SEQ ID NO: 9 is a first repetitive sequence of a plurality of repetitive sequences of SEQ ID NO: 9. 
     
     
       26. The vaccine of claim 21, in freeze-dried form. 
     
     
       27. The vaccine of claim 21, in the form of an injectable preparation. 
     
     
       28. The vaccine of claim 21, in a form suitable for injection, infusion or per os administration. 
     
     
       29. The vaccine of claim 21, further including an acceptable carrier. 
     
     
       30. The vaccine of claim 21, wherein the adjuvant includes Quil A, a cytokine, an oil and water emulsion, aluminum hydroxide, glucan, dextran sulfate, iron oxide, sodium alginate, poly amino acids, co-polymers of amino acids, saponin, paraffin oil, muramyl dipeptide, or CpG oligonucleotides. 
     
     
       31. The vaccine of claim 21, wherein the oil and water emulsion includes complete Freund's adjuvant or incomplete Freund's adjuvant. 
     
     
       32. The vaccine of claim 30, wherein the adjuvant includes Quil A. 
     
     
       33. An isolated polypeptide fragment of the polypeptide of SEQ ID NO: 4, having at least 15 contiguous amino acid residues of the amino acid sequence of SEQ ID NO: 4, which is effective for eliciting an immune response to Streptococcus or treating Streptococcus suis-associated diseases or infection caused by S. suis. 
     
     
       34. The isolated polypeptide fragment of claim 33, having at least 25 contiguous amino acid residues of the amino acid sequence of SEQ ID NO: 4. 
     
     
       35. The isolated polypeptide fragment of claim 33, having at least 35 contiguous amino acid residues of the amino acid sequence of SEQ ID NO: 4. 
     
     
       36. The isolated polypeptide fragment of claim 33, further including a fusion tag at the N-terminal end thereof. 
     
     
       37. The isolated polypeptide fragment of claim 33, further including at the C-terminal end thereof a repetitive sequence consisting of the amino acid sequence of SEQ ID NO: 9. 
     
     
       38. The isolated polypeptide fragment of claim 37, wherein said repetitive sequence is a first repetitive sequence of a plurality of said repetitive sequence. 
     
     
       39. The isolated polypeptide fragment of claim 34, further including at the C-terminal end thereof a repetitive sequence consisting of the amino acid sequence of SEQ ID NO: 9. 
     
     
       40. The isolated polypeptide fragment of claim 39, wherein said repetitive sequence is a first repetitive sequence of a plurality of said repetitive sequence. 
     
     
       41. The isolated polypeptide fragment of claim 35, further including at the C-terminal end thereof a repetitive sequence consisting of the amino acid sequence of SEQ ID NO: 9. 
     
     
       42. The isolated polypeptide fragment of claim 41, wherein said repetitive sequence is a first repetitive sequence of a plurality of said repetitive sequence. 
     
     
       43. The vaccine of claim 21, wherein the adjuvant includes carbohydrate, virosomes, structured complex of saponins and lipids, polymeric microsphere, killed bacteria, bacteria derivatives, toxin, peptidoglycan, lipopolysaccharide, trehalose dimycolate, or squalene.
