Abstract

The invention relates to pyrazole derivatives which are NMDA NR2B receptor inhibitors, useful in treating central nervous system diseases.

The invention claimed is: 
     
       1. A compound of Formula (I): 
       
         
           
           
               
               
           
         
       
       or a pharmaceutically acceptable salt, solvate, polymorph, or N-oxide thereof; wherein:
 n is 1 or 2, 
 Het is selected from: 
 
       
         
           
           
               
               
           
         
       
       wherein
 R 1  and R 2  are each independently selected from hydrogen; a halogen selected from F, Cl, and Br; C 1-6  alkyl, optionally substituted with at least one substituent, selected from hydroxy, halogen, C 1-3  (halo)alkoxy, and C 3-6  (halo)cycloalkyl; C 3-6  cycloalkyl, optionally substituted with at least one substituent, selected from hydroxy, halogen, C 1-3  (halo)alkyl and C 1-3  (halo)alkoxy; C 1-6  alkoxy, optionally substituted with at least one substituent, selected from hydroxy, halogen, C 1-3  (halo)alkoxy and C 3-6  (halo)cycloalkyl; —O—C 3-6  cycloalkyl, optionally substituted with at least one substituent, selected from hydroxy, halogen, C 1-3  (halo)alkyl and C 1-3  (halo)alkoxy; —S—C 1-3  alkyl, optionally substituted with at least one halogen atom; —SO 2 —C 1-3  alkyl, optionally substituted with at least one halogen atom; and —SF 5 ; or 
 R 1  and R 2  together form a 5 to 7 member carbocycle, optionally substituted with at least one substituent, selected from hydroxy, halogen, C 1-3  (halo)alkyl and C 1-3  (halo)alkoxy; and a 5 to 7 member heterocycle containing one to three heteroatoms which can be O, S or N, optionally substituted with at least one substituent, selected from hydroxy, halogen, C 1-3  (halo)alkyl and C 1-3  (halo)alkoxy; 
 R 3  and R 4  are each independently selected from hydrogen; halogen; C 1-5  (halo)alkyl, optionally substituted with at least one halogen atom, provided that at least one of R 3  and R 4  is hydrogen; 
 R 5  and R 6  are each independently selected from hydrogen; halogen; amino; C 1-5  alkyl, optionally substituted with at least one substituent, selected from halogen, hydroxy, C 1-3  (halo)alkoxy and C 3-6  (halo)cycloalkyl; C 1-5  alkoxy, optionally substituted with at least one substituent, selected from hydroxy, halogen, C 1-3  (halo)alkoxy, and C 3-6  (halo)cycloalkyl, and C 3-6  cycloalkyl, optionally substituted with at least one substituent, selected from hydroxy, halogen, C 1-3  (halo)alkyl and C 1-3  (halo)alkoxy; and 
 R 7  is hydrogen; C 1-5  alkyl, optionally substituted with at least one substituent, selected from halogen, hydroxy, C 1-3  (halo)alkoxy and C 3-6  (halo)cycloalkyl; or C 3-6  cycloalkyl, optionally substituted with at least one substituent, selected from hydroxy, halogen, C 1-3  (halo)alkyl and C 1-3  (halo)alkoxy, 
 wherein at least one of at least one of R 1  and R 2  is not hydrogen. 
 
     
     
       2. The compound of  claim 1 , wherein n is 1. 
     
     
       3. The compound of  claim 1 , wherein at least one of R 1  and R 2  is not hydrogen. 
     
     
       4. The compound of  claim 2 , wherein at least one of R 1  and R 2  is not hydrogen. 
     
     
       5. The compound of  claim 1 , wherein at least one of R 1  and R 2  is selected from F, Cl, Br, C 1-4  (halo)alkyl, C 3-6  (halo)cycloalkyl, C 1-3  (halo)alkoxy, C 1-3  (halo)alkoxy-C 1-3  (halo)alkyl, C 3-6  (halo)cycloalkyl-C 1-3  (halo)alkyl, and C 3-6  (halo)cycloalkyl-C 1-3  (halo)alkoxy and wherein R 3  and R 4  are each independently selected from hydrogen, F, Cl, Br, and C 1-3  (halo)alkyl, with the proviso that at least one of R 3  and R 4  is hydrogen. 
     
     
       6. The compound of  claim 1 , wherein R 3  and R 4  are each independently selected from hydrogen, F, Cl, Br, and C 1-3  (halo)alkyl, with the proviso that at least one of R 3  and R 4  is hydrogen. 
     
     
       7. The compound of  claim 1 , wherein:

 R 7  is hydrogen, C 1-3  alkyl, optionally substituted with at least one substituent, selected from halogen, hydroxy, C 1-3  (halo)alkoxy and C 3-6  (halo)cycloalkyl, or C 1-6  cycloalkyl, optionally substituted with at least one substituent, selected from hydroxy, halogen, C 1-3  (halo)alkyl and C 1-3  (halo)alkoxy.

 4-(3,4-dichlorophenyl)-2′-methyl-2′H-1,3′-bipyrazole, 
 4-(3-chloro-4-fluorophenyl)-2′-methyl-2′H-1,3′-bipyrazole, 
 2-((4-(4-chlorophenyl)-1H-pyrazol-1-yl)methyl)imidazo[1,2-a]pyridine, 
 3-(3-(difluoromethyl)-4-fluorophenyl)-1-((5-((4-(3-(difluoromethyl)-4-fluorophenyl)-1H-pyrazol-1-yl)methyl)-1H-pyrazol-3-yl)methyl)-1H-pyrazole, and

       12. A pharmaceutical composition comprising a therapeutically effective amount of a compound of  claim 1  or pharmaceutically acceptable salt, solvate, polymorph, or N-oxide thereof, and at least one pharmaceutically acceptable excipient. 
     
     
       13. The pharmaceutical composition of  claim 12 , further comprising a second active ingredient. 
     
     
       14. A method of treating a disease, disorder or condition associated with NMDA receptor hyperactivity comprising administering to a patient in need thereof a therapeutically effective amount of a compound of  claim 1 , or a pharmaceutically acceptable salt, solvate, polymorph, or N-oxide thereof. 
     
     
       15. The method of  claim 14 , wherein the disorder, disease or condition is selected from the group consisting of: bipolar disorder, major depressive disorder, and treatment-resistant depression. 
     
     
       16. A method for treating a disease, disorder or condition mediated by GluN2B receptors comprising administering to a patient in need thereof a therapeutically effective amount of a compound of  claim 1 , or a pharmaceutically acceptable salt, solvate, polymorph, or N-oxide thereof. 
     
     
       17. The method of  claim 16 , wherein the disorder, disease or condition is selected from the group consisting of treatment resistant depression and major depressive disorder. 
     
     
       18. A method of treating a disease, disorder or condition in a patient in need thereof comprising administering to the patient a therapeutically effective amount of a compound of  claim 1 , or a pharmaceutically acceptable salt, solvate, polymorph, or N-oxide thereof. 
     
     
       19. The method of  claim 18 , wherein the disease or disorder is a neurologic or psychiatric disorder. 
     
     
       20. The method of  claim 19 , wherein the disease or disorder is selected from the group consisting of a mood disorder; a stress-related disorder and neurodegeneration. 
     
     
       21. The method of  claim 20 , wherein the stress-related disorder is an anxiety disorder. 
     
     
       22. The method of  claim 19  wherein the neurologic disorder is epilepsy. 
     
     
       23. The method of  claim 17 , wherein the disorder, disease or condition is treatment resistant depression. 
     
     
       24. The method of  claim 17 , wherein the disorder, disease or condition is major depressive disorder. 
     
     
       25. The compound of  claim 1  selected from the group consisting of:

       26. A method of treating a disorder in a patient comprising administering to a patient in need thereof a compound of  claim 25  wherein the disorder is selected from the group consisting of bipolar disorder, major depressive disorder, and treatment-resistant depression. 
     
     
       27. A method of  claim 26  wherein the disorder is major depressive disorder. 
     
     
       28. A method of treating a disorder in a patient comprising administering to a patient in need thereof a compound of  claim 25  wherein the disorder is epilepsy.
